摘  要:目的:研究表皮特异性转录因子Ese-3在皮肤鳞状细胞癌中的表达及靶向沉默Ese-3基因表达对皮肤角质形成细胞Ha Ca T细胞增殖的影响。方法:免疫组化方法检测Ese-3在皮肤鳞状细胞癌及癌旁组织中的表达。利用慢病毒干扰系统感染Ha Ca T细胞,建立Ese-3基因敲减的Ha Ca T细胞系,MTT实验检测Ese-3敲除后细胞增殖的变化,流式细胞术检测Ese-3敲减后对细胞周期的影响。结果:免疫组化结果显示,Ese-3在皮肤鳞状细胞癌组织中表达显著低于癌旁皮肤组织。MTT和流式细胞术结果显示,下调Ese-3表达后,Ha Ca T细胞增殖加快,S期细胞增多,G0-G1期细胞和G2/M期细胞减少。结论:Ese-3在皮肤鳞状细胞癌中表达降低,抑制Ese-3基因表达可促进Ha Ca T细胞生长,提示Ese-3表达缺失可能通过促进细胞恶性增殖进而促进皮肤癌的发生发展。ObjectiveTo explore the expression of epidermal specific transcription factor Ese - 3 in squamous cell carcinoma of skin ( SCCS) and the function of Ese - 3 in the proliferation of HaCaT cell. Methods : Examine the ex-pression of Ese - 3 in squamous cell carcinoma of skin and para - carcinoma tissue by immunohistochemical staining. Using Ese - 3 interference lentivirus vector system to establish the cell model with Ese - 3 gene knocked down. MTT was used to detect cell growth. Flow cytometry was used to detect cell cycle. Results : Ese - 3 protein expression in SCCS tissue was significantly lower than in para - carcinoma tissue. Down regulation of Ese - 3 expression promoted HaCaT cells growth,and the percentage of the cells in phase S increased while in phase G0 - G2 and G2/M de-creased. Conclusion : The expression of Ese -3 decreased in squamous carcinoma tissue. Down regulation of Ese -3 expression can promote HaCaT cells growth and the development of SCCS.
