下调OTX1的表达抑制直肠癌Colo320细胞的增殖和侵袭

摘　　要：目的:下调直肠癌细胞系Colo320的Orthodenticle人同源盒基因1(orthodenticle homeobox 1,OTX1)的表达,观察其对Colo320细胞增殖和侵袭能力的影响。方法:构建OTX1 siRNA重组质粒载体p GPU6-OTX1,并转染Colo320细胞。应用qRT-PCR法检测OTX1的基因表达水平,Western blot法检测OTX1的蛋白表达,CCK-8法检测p GPU6-OTX1对Colo320细胞增殖能力的影响,Transwell小室法检测Colo320细胞侵袭能力的变化。结果:成功将构建的p GPU6-OTX1转染Colo320细胞。qRT-PCR和Western blot结果显示p GPU6-OTX1下调Colo320细胞OTX1的表达,CCK-8结果显示p GPU6-OTX1明显抑制SGC-7901细胞的增殖能力,Transwell结果显示下调OTX1的表达抑制Colo320细胞的侵袭能力。结论:下调OTX1的表达能够抑制直肠癌细胞Colo320的增殖和侵袭能力。ObjectiveDown - regulation the orthodenticle homeobox 1 （0T X1 ） in rectal cancer Colo 320 ce lls,to investigate its effects on proliferation and invasion. Methods ： Construction of OTX1 siRNA plasmid pGPU6 - OTX1, transfected into Colo 320 cells. The OTX1 gene express was detected by quantitative real - time PCR,the OTX1 protein expression was detected by Western blot,the proliferation of Colo 320 cells was analyzed by CCK - 8 assay,the inva-sion of Colo 320 was detected by Transwell. Results： The pGPU6 - OTX1 was successfully constructed and transfected into Colo 320 cells.The result of real-t ime quantitative PCR and Western blot were showed that the OTX1 was down - regulated by pGPU6 - OTX1. The proliferation and invasion were significantly inhibited in Colo 320 cells by pGPU6 - OTX1. Conclusion ： Down - regulation the expression of OTX1 inhibits cell proliferation and invasion in rectal cancer Colo 320 cells.
