摘  要:目的建立枇杷花水提液中总酚的含量测定方法,优化总酚测定的最佳条件。方法以没食子酸为对照品,在760nm处采用Folin-Ciocalteu法测定总酚的含量,考察福林-酚用量、Na_2CO_3溶液用量、显色时间等对测定的影响,并建立最佳测定条件。结果 Folin-Ciocalteu法测定枇杷花水提液中总酚,最佳福林-酚用量为3ml,最佳10%Na_2CO_3溶液用量为6ml,最佳显色时间为50min,多酚质量浓度在0.4~3.2μg/ml范围内与吸光度线性关系良好,其回归方程为Y=0.1419X+0.1427,R^2=0.9974;加样回收率为96.19%,RSD为2.03%,该测定方法精密度、重复性和稳定性良好。结论该方法简便易行,稳定性好,对提高枇杷花水提液的质量控制标准具有参考意义。Objective To establish a rapid determination of total phenols in water extraction of flowers of Eriobotrya japonica ( Thunb. ) Lindl and optimize the reaction condition. Methods The content of total phenols was determined at 760nm by Folin - Ciocalteu colorimetric method. Gallic acid was used as the control and the single factor tests were adopted to screen chromogenic conditions, such as the amount of Folin - Ciocalteu reagent, the amount of sodium carbonate solution and the dark reaction time. Results The optimal chromogenic conditions were as follows: the amount of Folin - Ciocaheu reagent 3mL, the amount of sodium carbonate solution 6 mL and the dark reaction time 50 min. Under the optimum conditions, the mass concentration of polyphenols in the range of 0.4 - 3.2μg/mL had a good linear relationship between absorbance (y -- 0. 1419x +0. 1427) with the correlation coefficient of 0. 9995. The average recovery was 96.19% and RSD was 2.03% (n = 6). This method has good precision , repro- ducibility and stability. Conclusion This method is simple, stable and useful for enhancing the quality control standards of water extract of flowers of Eriobotrya japonica ( Thunb. ) Lindl.
