摘 要:目的:探讨瘢痕灸预处理肝俞及足三里穴对原发性肝癌大鼠Wnt信号传导通路相关蛋白β-连环蛋白(β-catenin)、分泌型卷曲相关蛋白(s FRP1)表达的影响。方法:48只Wistar雄性大鼠,随机分为对照组、模型组、预处理肝俞组、预处理足三里组,每组12只。预处理肝俞组和预处理足三里组均提前2周施行瘢痕灸后,与模型组一同采用腹腔注射二乙基亚硝胺(50 mg/kg)建立大鼠原发性肝癌(primary liver cancer,PLC)模型,记录各组大鼠体质量,造模结束后取大鼠肝脏,采用Western blot方法测定β-catenin,s FRP1表达量,HE染色观察大鼠肝脏组织形态学变化。结果:与对照组相比,模型组、预处理肝俞组和预处理足三里组的s FRP1表达量明显下降(P<0.05),β-catenin的表达量明显升高(P<0.05);与模型组比较,预处理肝俞组和预处理足三里组s FRP1表达量明显升高(P<0.05),β-catenin的表达量显著降低(P<0.05);与预处理肝俞组比较,预处理足三里组s FRP1表达量明显降低(P<0.05),β-catenin的表达量升高(P<0.05)。结论:瘢痕灸预处理肝俞和足三里对预防原发性肝癌均有良性作用,且肝俞穴效果优于足三里穴。Objective: To explore the effects of scarring moxibustion pretreatment Ganshu (BL 18) and Zusanli( ST 36) on Wnt signaling related protein β - catenin and sFRP1 expression pathway of primary liver cancer in rats. Methods : A total of 48 Wistar rats were randomly divided into the control group, model group, moxibustion preconditioning at Ga shu group and moxibustion preconditioning at Zusanli group, with 12 rats in each group. Both moxibustion preconditioning at Ganshu and Zusanli groups were the purposes of scarfing moxibustion before two weeks, and together with the model group by intraperitoneal injection of diethylnitrosamine (50 mg/kg) to establish rat primary liver cancer (PLC) model, and recording the weight of rats in each group. The liver was removed after ending rat model, and expression of β - catenin and secreted frizzled - related protein ( sFRP1 ) were measured by Western blot, and at the end of model establishment, observe the liver morphology change with HE staining. Results: Compared with the control group, the protein expressions of sFRP1 in the model group, moxibustion preconditioning at Ganshu group and moxibustion preconditioning at Zusanli group were decreased significantly ( P 〈 0. 05 ), and β - catenin expression was increased ( P 〈 0.05 ). Compared with the model group, the sFRP1 expression of moxibustion preconditioning at Ganshu group and moxibustion preconditioning at Zusanli group was significantly higher ( P 〈 0. 05 ) and the expression of β - catenin was decreased ( P 〈 0. 05 ). Compared with moxibustion preconditioning at Ganshu group, the sFRP1 expression was lower and the β - cate- nin expression was higher in the rnoxibustion preconditioning at Zusanli group ( P 〈 0. 05 ). Conclusion. Pretreatment scarring moxibustion on Ganshu and Zusanli of rats to prevent primat7 liver cancer has a benign effect, and the effect at Ganshu is better than Zusanli.
