摘  要:目的:研究虫草素在体外对舌癌TCA-8113细胞周期和凋亡的影响;方法:以噻唑蓝法(MTT)测定虫草素对TCA-8113细胞增殖活性的影响;用流式细胞术测定虫草素对TCA-8113细胞周期的影响情况以及是否诱导TCA-8113发生凋亡作用;采用蛋白免疫印迹检测细胞周期相关蛋白和细胞凋亡相关蛋白的表达情况;结果:虫草素对TCA-8113细胞增殖活性的抑制作用具有浓度依赖性,能够引起TCA-8113细胞发生G2期细胞周期阻滞,并诱导TCA-8113细胞发生凋亡作用;Cyclin B1和Bcl-2表达减少,Cdc2和Bax表达量没用明显变化。结论:虫草素有效的引起TCA-8113发生细胞周期阻滞,诱导TCA-8113细胞凋亡,从而抑制TCA-8113细胞的增殖活性,为进一步应用于临床治疗舌癌等肿瘤疾病提供了依据。Objective :To observe the effect of cordycepin on cell cycle arrest and apoptosis in TCA -8113 cell line in vitro. Methods:The effect of cordycepin on proliferation of TCA -8113 cells was determined by MTT method. The apopto- sis and the cell cycle arrest of TCA -8113 cells were determined by flow cytometry with FITC -Annexin V and PI. The proteins of Cyclin B1, Cdc2, Bcl - 2 and Bax were determined by Western blot. Results : Cordycepin showed a dose - de- pendent inhibitory effect on the proliferation of TCA- 8113 cells. It induced the apoptosis of TCA -8113 cells and cell cycle arrest at G2 phase in dose- dependent manner. The CyctinB1 and Bcl -2 decreased in TCA -8113 with the in- creasing concentrations of cordycepin. Cdc2 and Bax had no obvious change. Conclusion:Cordycepin can cause TCA - 8113 cell cycle arrest and apoptosis by decreasing CyclinB1 and Bcl- 2 expression in vitro.
