摘  要:以菠菜、油菜、小麦、冬青叶为实验材料,应用分光光度法测定叶绿素提取量,并采用正交法进行条件优化。结果表明:不同因素对不同植物材料叶绿素提取量的影响不同。小麦、冬青叶绿素的提取量最高时,其处理温度均为40℃,提取液均为丙酮∶乙醇=1∶1,处理时间分别为36 h、12 h;叶绿素的提取量最高时,菠菜的处理温度为60℃,处理时间为1.5 h,提取液为乙醇;而油菜处理温度为50℃,处理时间为4.5 h,提取液为丙酮∶乙醇=1∶2。波菜可作为此实验项目中较适合的教学材料。The extraction of chlorophyll was guided by spectrophotography, and the extraction conditions were optimized by orthogonal experiment with spinach, rape, wheat and holly leaves as experimental materials. The results showed that the influence of different factors on the chlorophyll extraction of different plant materials was different. The highest extraction of chlorophyll in wheat and holly leaves was as follows: the extraction temperature was 40 ~C, the extract was acetone : ethanol = 1 : 1, the extraction time was 36 h and 12 h, respectively. The highest extraction of chlorophyll in spinach leaves was as follows: the extraction temperature was 60 ℃, the extraction time was 1.5 h, the extract was ethanol. The highest extraction of chlorophyll in rape leaves was as follows: the extraction temperature was 50 ℃, the extraction time was 4.5 h, the extract was acetone : ethanol = 1 : 2. As the result, spinach was the suitable material in the experimental item.
