摘 要:目的探讨Cultured ELISPOT试验检测肿瘤-睾丸抗原特异性CTL反应的能力。方法选择2016年1月-6月新疆医科大学附属肿瘤医院头颈放疗科确诊头颈部鳞癌肿瘤住院患者26例,采用ELISPOT试验(ELISPOT组),选择2016年10月-2017年2月新疆医科大学附属肿瘤医院头颈放疗科确诊头颈部鳞癌肿瘤住院患者17例,采用Cultured ELISPOT试验(Cultured ELISPOT组)。用合成的肿瘤-睾丸抗原(CTAs)NYESO1、SSX2、SALL4、MAGEA1、MAGEA3特异性CTL抗原表位肽刺激患者外周血单个核细胞(PBMC),分别检测两组患者分泌IFN-γ的CTL细胞的频率及强度,并比较两组差异。结果 (1)Cultured ELISPOT组患者特异性CTL平均反应频率高于ELISPOT组患者,差异无统计学意义(P>0.05)。(2)Cultured ELISPOT组患者平均反应强度明显高于ELISPOT组患者,差异均有统计学意义(P<0.05),且Cultured ELISPOT组患者平均反应强度至少高于ELISPOT组患者25倍以上。结论 Cultured ELISPOT试验可以部分提高特异性T细胞免疫反应的平均反应频率及强度,有助于T细胞免疫在肿瘤中的研究。Objective To investigate the capacity of testing the immune response to specific cytotoxic lym-phocyte induced by cancer-testis antigen by Cultured ELISPOT. Methods 26 head and neck squamous cell carcinomas(HNSCC) from January 2016 to June 2016 were enrolled into the study from Xinjiang Tumor hospital subjected to ELISPOT (ELISPOT group). 17 HNSCC from October 2016 to February 2017 were enrolled from Xinjiang Tumor hospital preparing to use Cultured ELISPOT (Cultured ELISPOT group). The cancer-testis antigen (CTAs) NYESOl,SSX2 , SALL4, MAGEA1 , MAGEA3 overlapping peptides were synthesized to use. The frequency and magnitude of specific cytotoxic immune response secreting IFN-y were observed by ELISPOT and Cultured ELISPOT respectively. Results (1) The Cultured ELIS-POT group whose CTL average reaction frequency were shown to be higher than that in the ELISPOT group, the difference were not significant. (2) The magnitude of Cultured ELISPOT group was shown to be higher than that in ELISPOT group however, no sigfnificant difference was observed. Conclusion Cul-tured ELISPOT may improve the immune response to specific CTL, which could facilitate the research of T cell immune in tumor.
