摘　　要：目的优化测定癫痫患儿血清中丙戊酸钠浓度的方法。方法以环己烷羧酸作为内标,血样经乙腈蛋白沉淀和酸化后,以低浓度2-溴-对硝基苯乙酮为衍生化试剂,三乙胺为催化剂,在超声辐射下进行衍生化反应。色谱柱为Bennetnach C_(18)柱,流动相为甲醇-水(79∶21),柱温:25℃,流速:为1ml·min^(-1),检测波长:265nm。结果丙戊酸钠血药浓度在5~160μg·ml^(-1)范围内与峰面积呈良好线性关系(r=0.9995),平均回收率为94.8%,日内与日间RSD≤8.7%。结论本法可简便、快速、准确地测定癫痫患儿血清中丙戊酸钠浓度,适用于临床常规监测需要。Objective To improve an HPLC method for determination of sodium valproate in the serum of epileptic childern.Methods Acetonnitrile was used as the protein precipitant.Serum samples were extracted with hexane.A low concentration of 2-bromine-p-nitrophenyl ethyl ketone was selected as the derivative reagent under uitrasonic radiation,with triethylamine as the catalyst.A UV detector（265nm）and Bennetnach C18 column were used,and temperature of the column was 25℃.The mobile phase consisted of methanol alcohol-water（79∶21）at a flow rate of 1.0ml·min^-1.Results The linear range of valproic acid was 5-160μg·ml^-1（r=0.9995）.The mean recovery was 94.8%.The inter-day and intra-day RSD was less than 8.7%.Conclusion This method is simple,accurate,specific and applicable to clinical routine monitoring.
