摘 要:【目的】分析菜心-小菜蛾互作的生理和蛋白组变化规律,为明确菜心对小菜蛾取食的抗虫信号途径及防御机理提供理论依据。【方法】以四九菜心四叶幼苗为材料,测定菜心叶片接入小菜蛾24 h前后的碳水化合物代谢变化,并应用串联质量标签(tandem mass tag,TMT)标记联合高效液相色谱-质谱/质谱法(LCESI-MS/MS)技术鉴定菜心接入小菜蛾24 h前后的差异蛋白组变化。【结果】菜心接虫后较对照可溶性淀粉积累减少,可溶性蛋白及可溶性糖(包括蔗糖、还原糖)含量增加;本研究共筛选出差异表达蛋白430个(接虫后较对照上调1.2倍以上的蛋白211个,下调0.83倍以下的蛋白119个)。其中与光合作用合成代谢相关的许多蛋白在小菜蛾取食菜心后下调表达,而与逆境胁迫、糖水解及能量代谢相关的一些蛋白上调表达。【结论】TMT技术能有效地筛选出小菜蛾取食菜心前后的差异表达蛋白,其差异蛋白主要参与生物胁迫、糖代谢、光合作用和呼吸作用的能量及物质的转化等生物过程。[ Objective] The changes of carbohydrate metabolism and expression of proteins were analyzed to provide the basic theory information for exploring the interactions between the diamondback moth feeding and defense response of Chinese flowering cabbage. [ Method] After 24 h the diamondback moth feeding on ' Sijiu' Chinese flowering cabbage, the study analyzed rules for soluble starch, soluble protein, sugar and reducing sugar, meanwhile, analyzed expression of proteins by Tandem mass tag (TMT) coupled to two-dimensional liquid chromatography tandem mass spectrometry (2D LC-MS/MS). [ Result] The soluble starch accumulation decreased, and soluble protein and soluble sugar (inchiding sugar, reducing sugar) content increased. 430 discrepant proteins were detected by bioinformatics analysis using the SEQUEST search engine, 211 proteins were up-regulated with a more than 1.2 folds change, and 119 proteins were down-regulated with a less than O. 83 folds. Among them, the ones of photosynthesis were down regulated, while the others related to stress response, some of hydrolytic en- zymes and energy metabolism were up regulated. [ Conclusion ] The TMT was a powerful method for isolating and identifying differentially expressed proteins between the diamondback moth feeding and defense response of Chinese flowering cabbage. Most of them were involved in stress resistance, glycometabolism, photosynthesis, respiration, energy and substance metabolism.
