摘 要:目的探讨sn-2棕榈酸酯(OPO)结构脂肪对低钙大鼠钙吸收及骨量的影响。方法断乳SD大鼠24只分为三组,以正常AIN-93G饲料为对照组,低钙饲料为骨质流失模型组,在低钙饲料中添加57.0%OPO结构脂肪为OPO组(Ca和OPO相对含量:0.5%和10.4%、0.1%和10.4%、0.1%和57.0%)。6w后进行钙吸收实验。12w后测定血钙水平、肠粘膜钙转运基因相对表达量、全身和脊柱骨密度(BMD)和骨矿量(BMC),观察左胫骨骨微结构。结果与对照组比较,模型组大鼠最终身长短小、骨量降低、骨小梁厚度减小(P<0.05),表明低钙膳食成功诱导骨质丢失模型;与模型组比较,OPO组大鼠血钙水平略见升高[(2.54±0.09)vs(2.36±0.26)mmol/L,P=0.0427],钙转运相关蛋白各基因表达显著上升(P<0.01),大鼠身长明显增长[(25.13±0.99)vs(23.63±0.92)cm,P=0.007],但骨密度、骨矿量及骨微结构未见明显改善(P>0.05)。结论 57.0%OPO通过上调钙转运基因表达促进钙的吸收利用,可能改善大鼠由于低钙引起的生长发育不良。Objective To investigate the effects of beta-palmitate (OPO) on calcium absorption and bone mass in calcium-deficient rats. Methods Twenty-four weaning SD rats were randomly assigned to three groups of equal size: control group fed a normal diet, model group given with a low calcium diet and OPO group consuming a low calcium diet with OPO (Ca% and OPO% in each group: 0.5% and 10.4%, 0.1% and 10.4%, 0.1% and 57.0%, respectively). After 6w of feeding, rats were individually housed 3 days for calcium balance study. After 12 w of feeding, the levels of serum calcium, expressions of calcium transport genes, whole-body and spine bone mineral density (BMD), bone mineral content (BMC), and microarchitecture of left tibia were measured. Results Compared with the control group, final body length, bone mass and trabecular thickness were significantly decreased in model group (P〈0.05). Supplementation of 57.0% OPO increased serum calcium level, expression of calcium transport genes, as well as body length compared with those in model group (P〈0.05), but with no significant difference in BMD, BMC and bone microarchitecture. Conclusion Supplementation of 57.0% OPO is effective in increasing calcium absorption by up-regulating calcium transport genes expression, and probably improve the compromised body growth induced by calcium deficiency.
