摘 要:本文通过TNBS法、OPA法、茚三酮比色法三种方法对革屑水解物、乳清蛋白水解物和毛发水解物的水解度进行了测定。结果表明:三种方法在测量革屑水解物和乳清蛋白水解物的水解度时结果比较接近,茚三酮法和OPA法略低;但在测量毛发水解物时OPA法的测量结果明显小于TNBS法和茚三酮法;同时茚三酮法吸光度波动较大,不稳定;OPA法和TNBS法的测定结果更能接近实际的游离氨基浓度,从而水解度的计算更准确,而且OPA法操作更简便,反应时间更短。In this paper, the degree of hydrolysis of the leather scraps hydrolysate,whey protein hydrolysate and hair hydrolysate were determined by TNBS, OPA, ninhydrin colorimetry. The results showed that:The results of these three methods were relatively close in measuring the degree of hydrolysis of the leather scraps hydrolysate and whey protein hydrolysate,Indene three ketone and OPA slightly lower; but the measurement resuhs of OPA method in the measurement of hair hydrolysate was significantly less than the TNBS method and three indene ketone method; at the same time, Yin three ketone absorbance fluctuated widely, unstable; the results of OPA method and TNBS method were more close to the actual concentration of free amino acid, so the calculation of degree of hydrolysis was more accurate, and the OPA method was more simple and convenient, and the reaction time was shorter.
