摘  要:目的:为了研究长链非编码RNA(lnc RNA)在益髓生血颗粒治疗前后患者红细胞(含网织红细胞)和外周血单个核细胞(PBMC)中表达谱的变化,并分析二者的差异。方法:利用lnc RNA芯片技术分别检测3例中间型β-地贫患者益髓生血颗粒治疗前后患者红细胞(含网织红细胞)和PBMC表达谱,通过对原始数据进行预处理达到均一化后,筛选出差异表达lnc RNA并进行分析。结果:与用药前的lnc RNA表达比较,红细胞中变化2倍以上lnc RNA共6条,PBMC中变化2倍以上且P<0.05的lnc RNA共260条。结论:益髓生血颗粒治疗前后,患者红细胞和PBMC中的lnc RNA表达谱存在差异,为进一步揭示中医肾藏精生髓、髓生血治疗地中海贫血理论内涵打下基础。Objective:To analyze the expression profile of long non-coding RNA(lnc RNA)in erythrocyte including reticulocytes and peripheral blood mononuclear cell(PBMC)before and after treatment of Yisui Shengxue Granule. Methods:The technique of lnc RNA microarray was used to inspect the lnc RNA expression profile in tin erythrocyte including reticulocytes and peripheral blood mononuclear cell before and after treatment of Yisui Shengxue Granule. The raw data of lnc RNA were pretreated for normalization. Results:Through the analysis,the research says that compared with the lnc RNA expression before treatment,6 lnc RNAs which had more than 2-fold variation in erythrocyte,while 260 lnc RNAs had more than 2-fold variationand significant difference(P〈0.05)by statistical analysis were regarded as lnc RNA with differentialexpression. Conclusion:Lnc RNA expression profiles of before and after treatment of Yisui Shengxue Granule change significantly in erythrocyte including reticulocytes and PBMC of intermediate type of β-thalassemia,peripheral blood mononuclear cell before and after treatment. This study laid the foundation of reveal the theoretical connotation of"kidney producing marrow,marrow producing blood".
