摘 要:目的探讨不同的诱导液对牙周膜干细胞(PDLSCs)膜片生物学活性的影响。方法分别使用骨髓间充质细胞条件(BMMSCs-CM)诱导液与根端牙胚条件(APTG-CM)诱导液培养人PDLSCs,并制备成PDLSCs膜片。BMSCs-CM诱导液诱导的细胞设为BMSCs-CM组,APTG-CM诱导液诱导的细胞设为APTG-CM组。分别诱导PDLSCs膜片4、7、10 d。通过倒置显微镜、HE染色、免疫组化、扫描电镜检测并观察两种诱导液诱导后的PDLSCs细胞膜片形态学的变化及生物学特性的差异。结果接种10 d后,BMSCs-CM组和APTG-CM组的克隆形成率分别为37%和24%,BMSCs-CM的克隆形成能力明显强于APTG-CM组,差异有统计学意义(P<0.05)。与APTG-CM诱导液比较,经过BMMSCs诱导液诱导后的PDLSCs膜片可产生更丰富的细胞外基质。扫描电镜可见BMMSCs诱导液诱导后的细胞膜片表面有矿化沉积和胶原组织形成;APTG-CM诱导液诱导后的细胞膜片表面有大量胶原纤维状组织形成。结论 BMMSCs诱导液更适合诱导PDLSCs膜片向类牙周组织分化。Objective To explore the change of membrane biological activity of PDLSCs under the different conditions of induced liquid. Methods Respectively cultivated BMMSCs-CM and APTG-CM to be cultured people PDLSCs,and prepared into PDLSCs membrane. Cells induced by BMSCs-CM were in BMSCs-CM group, the others induced by APTG-CM were in APTG-CM group. Respective- ly induced PDLSCs membrane with two kinds of culture solutions for 4,7 and 10 days. Through inverted microscope,HE dyeing,IHC, SEM test and observed the PDLSCs cell membrane morphology change and biological characteristics difference induced by different induced liquid. Results After 10 days of inoculate,cloning efficiency was 37% in BMSCs-CM group and 24% in APTG-CM group, clonality in BMSCs-CM group was significantly stronger than that in APTG-CM group, the difference has statistically significance(P 〈 0. 05). Compared with APTG-CM induced liquid, the membrane induced by BMMSCs induced liquid could generate more plentiful ECM. It was found that there were mineralized collagen deposition and collagenous tissue forming on membrane surface induced by BMMSCs through SEM, and there were large numbers of collagen fibrous shape tissue forming on membrane surface induced by APTG- CM. Conclusion Compared with APTG-CM induced liquid, BMMSCs induced liquid is fitter to induce the differentiate type periodontal tissue differentiation of PDLSCs membrane.
