摘 要:目的探讨氧糖剥夺再灌注(oxygen-glucose deprivation/reperfusion,OGD/R)后SH-SY5Y细胞凋亡情况及NF-κB信号通路NF-κBp65、COX-2蛋白的表达。方法将SH-SY5Y细胞分为正常对照组、氧糖剥夺4 h/再灌注24 h组(OGD/R)组。利用流式细胞术分别检测各组细胞凋亡率,蛋白印记法(Western blot)检测p65和COX-2的蛋白表达情况。结果与正常对照组比较,OGD/R组细胞凋亡率明显增高(P<0.05),p65和COX-2的蛋白表达也明显增高(P<0.05)。结论氧糖剥夺4 h/再灌注24 h能够促进细胞凋亡,其机制可能是通过NF-κBp65对COX-2调控的信号通路来实现。Objective To explore the relationship between the apoptosis of SH-SY5Y cells and NF-KBp65 , COX-2 after oxygen-glucose deprivation/reperfusion (OGD/R). Methods Cells were divided into normal control group and oxygen glucose deprivation 4 h/reperfusion 24 h groups. The cell apoptosis rate was analyzed using flowcytometry, and Western blot assay was used to evaluate the expression of p65 and COX-2 proteins in control group and OGD/R group. Results Compared with the normal control group,the apoptosis rate of OGD/R group was significantly higher (P 〈 0 .0 5 ) ,and the protein expres-sion of p65 and COX-2 was also significantly increased (P 〈 0 .0 5 ) . Conclusion Oxygen glucose deprivation 4 h/reperfusion 24 h can induced apoptosis,which may be through the signaling pathway that NF-kB p65 regulates COX-2 to achieve.
