SIRT3在乳腺癌小鼠模型中的表达及其临床意义的研究

摘　　要：目的探讨乳腺癌小鼠模型中SIRT3蛋白的表达水平及其临床意义。方法通过免疫组化和蛋白质印迹法(Western Blot)检测SIRT3蛋白在小鼠乳腺肿瘤组织及乳腺癌细胞中的表达情况,通过实时荧光定量PCR检测SIRT3 mRNA在小鼠乳腺肿瘤组织中的表达情况。结果免疫组化结果显示,SIRT3在乳腺肿瘤组织中的阳性表达率[(17.17±2.08)%]明显低于正常组织[(33.33±2.65)%],差异有统计学意义(P﹤0.01)。Western Blot结果显示,SIRT3在乳腺癌细胞中的相对表达量(0.220±0.173)明显低于正常细胞(0.850±0.086),差异有统计学意义(P﹤0.01)。实时荧光定量PCR结果显示,SIRT3 mRNA在乳腺肿瘤组织中的表达水平(0.520±0.069)明显低于正常组织(0.886±0.034),差异有统计学意义(P﹤0.01)。结论乳腺肿瘤组织和乳腺癌细胞中SIRT3表达下调,SIRT3可能在乳腺癌的发生、发展过程中起关键作用。Objective To investigate the expression of SIRT3 in mouse model of breast cancer and to explore its clini- cal significance. Method Immunohistochemistry and Western Blot were used to detect the expression of SIRT3 in breast cancer tissues and breast cancer cells, and SIRT3 mRNA expression by real-time PCR. Result The immunohistochemis- try showed that the positive expression rate of SIRT3 protein in breast cancer tissue [（17.17±2.08）%] was obviously lower than that in normal tissues [（33.33±2.65）%] （P〈0.01）. The Western Blot showed that the relative expression rate of SIRT3 in breast cancer cells （0.220±0.173） was lower than that in normal cells （0.850±0.086） （P〈0.01）. And similar results were observed for SIRT3 mRNA in breast cancer tissues （0.520±0.069） and normal tissues （0.886±0.034） （P〈0.01）. Conclusion SIRT3 expression was down-regulated in breast cancer tissues and cells, indicating a crucial role of SIRT3 in the develop- ment of breast cancer.
