摘  要:目的探讨维吾尔族和汉族子宫颈癌(CC)患者中肿瘤干细胞相关基因POU5F1和NANOG在mRNA水平表达的差异及其与临床病理的关系。方法采用实时荧光定量PCR法(RT-PCR),检测69例子宫颈癌组织和16例子宫颈癌组织及其相应的癌旁新鲜组织标本中POU5F1、NANOG在mRNA水平上的表达情况,了解其在维吾尔族和汉族间的表达差异。根据患者临床分期、肿瘤分化程度、淋巴结转移进行分组,比较不同亚组患者中维吾尔族和汉族患者间POU5F1、NANOG表达水平的差异性。结果子宫颈癌组织中POU5F1 mRNA、NANOG mRNA的表达水平均较癌旁组织高(P﹤0.05)。维吾尔族CC患者POU5F1 mRNA、NANOG mRNA表达水平均高于汉族CC患者(P﹤0.05)。临床分期为Ⅱa~Ⅲ期、淋巴结转移阴性时,维吾尔族与汉族CC患者子宫颈癌组织中POU5F1 mRNA表达水平比较,差异有统计学意义(P﹤0.05);临床分期为Ⅱa~Ⅲ期、中-高分化、淋巴结转移阴性时,维吾尔族与汉族CC患者子宫颈癌组织中NANOG mRNA表达水平比较,差异有统计学意义(P﹤0.05)。结论 CC中肿瘤干细胞相关基因POU5F1 mRNA和NANOG mRNA的表达在维吾尔族和汉族人群中存在差异性,而此种差异性在Ⅱa~Ⅲ期、中高分化、淋巴结转移阴性的CC中表现更加明显。Objective To investigate the differences in expression levels of tumor-stem-cell-related gene POU5F1 and NANOG at mRNA level between Xinjiang Uyghur and Hart patients with cervical cancer, as well as its effects on clin- icopathologic results. Method The expression of POUSF1 and NANOG at mRNA level in 69 cervical cancer tissues and 16 cervical cancer tissues plus their matching flash para-carcinoma tissues were detected by real-time fluorescent quantita- tive PCR (RT-PCR), to analyze the differences in their expression levels between Uyghur and Han ethnic groups. Patents were divided into subgroups according their clinical stages, degree of tumor differentiation and lymphatic metastasis to examine the differences in expression levels of POU5F1 and NANOG between Uyghur and Han patients in different sub- groups. Result The expression levels ofPOU5F1 mRNA and NANOG mRNA in cervical cancer tissues were both high- er than those in para-carcinoma tissues (P〈0.05). The expression levels of POU5F1 mRNA and NANOG mRNA in Ui- ghur patients with cervical cancer were both higher than those in their Han counterparts (P〈0.05). When comparing in stage Ⅱa-Ⅲ and lymph node negative subgroup, difference in the expression level of POU5F1 mRNA between Uyghur and Han patients with cervical cancer reached statistical significance (P〈0.05); when comparing in stage IIa-III, moderate- ly to highly differentiated and lymph node negative subgroup, difference in the expression level of NANOG mRNA be- tween Uyghur and Han patients with cervical cancer reached statistical significance (P〈0.05). Conclusion The expres- sion levels of tumor-stem-cell-related gene POU5F1 and NANOG at mR_NA level in Xinjiang Uyghur patients with cervi- cal cancer are different from those in their Han counterparts, and the differences are more significant in stage Ⅱa-Ⅲ, mod- erately to highly differentiated and lymph node negative patients with cervical cancer.
