摘 要:目的:观察局灶性脑梗死大鼠梗死区大脑皮质过氧化物酶6(peroxiredox6,PRX6)表达和丙二醛(malondialdehyde,MDA)含量的变化。方法:30只雄性SD大鼠随机分为对照组和模型组,采用线栓法制作大鼠大脑中动脉栓塞模型,各组大鼠分别于术后1d、3d、7d处死取材。采用硫代巴比妥酸法检测梗死区大脑皮质MDA的含量,Western blot法和q RT-PCR法检测梗死区大脑皮质PRX6蛋白和mRNA的表达。结果:模型组各时间点大鼠梗死区大脑皮质MDA含量,PRX6蛋白、mRNA表达均明显高于对照组(P<0.01)。随着术后时间的延长,模型组大鼠梗死区大脑皮质MDA含量逐渐降低,PRX6蛋白、mRNA表达逐渐升高(P<0.01)。结论:大鼠脑梗死后可能通过上调PRX6的表达水平减轻氧化应激损伤。Objective: To investigate the changes of peroxiredox6 (PRX6) expression and malondialdehyde (MDA) content in cerebral cortex of rots after local cerebral infarction. Methods: 30 male SD mrs were randomly divided into control group and model group. Suture method was used to establish middle cerebral artery occlusion rats' model, and the rats were killed respectively ld, 3d, 7d after operatiorL Thiobarbituric acid method was used to detect lfie MDA content in cerebral cortex, Western blot and qRT-PCR to detect PRX6 protein and mRNA expression in cerebral cortex. Results: The MDA content and PRX6 expression in infarct area of cerebral cortex of rats in model ~oup at each time point were obviously higher than control group (P〈0.01). With time extension after operation, the MDA content declined gradually, while the PRX6 expression increased gradually (P〈 0,01 ). Conclusions: After cerebral farction, the rats may reduce oxidative stress injury by upregulafing PRX6 expression.
