摘 要:为研究柠檬酸合成酶基因与‘琯溪蜜柚’果实"返酸"的关系,以不同贮藏期‘琯溪蜜柚’果实汁胞为材料,克隆了1个柠檬酸合成酶基因,将其命名为CmCS,其含有1 416 bp开放阅读框,编码具有471个氨基酸的蛋白。生物信息学分析表明,CmCS推导的氨基酸序列与甜橙、川桑等植物的相似度皆大于90%,为稳定的亲水蛋白,亚细胞定位预测其定位于线粒体。结果表明,‘琯溪蜜柚’贮藏期间伴随着有机酸含量逐渐增加,CmCS基因表达量整体先升高后降低,推测其对‘琯溪蜜柚’果实采后有机酸代谢具有一定调控作用。In order to study the relationship between the citrate synthase gene and the " sour regurgitation" of ‘ Guanximiyou’ fruit.A citrate synthase gene named CmCS was cloned by PCR from ‘ Guanximiyou’ fruits,which stored in different days of storage.The open reading frame of CmCS was 1 416 bp,encoding a protein of 471 amino acids.Bioinformatics analysis showed that the amino acid sequence was greater than 90% homology with Citrus sinensis and Morus notabilis.Sequence analyses revealed that this protein was stable and hydrophilic,the subcellular localization was in the mitochondria.And the relative expression of this gene was surveyed by real-time quantitative PCR during different days of storage.The results showed that,its relative expression during different days of storage was different;the CmCS expression was low in the early days,and expression gradually increased then reduced,With the increase of organic acid content.In conclusion,it suggested that CmCS could play a role for regulation of organic acid metabolism in ‘ Guanximiyou’ fruits.
