摘 要:为明确H 027×陇薯6号和H-027×陇薯7号2个杂交组合杂种F1的4个优异新品系(A 2、A 10、B 1和B 12)在DNA水平上的遗传差异,利用SSR分子标记技术对4个新品系进行多态性分析。利用筛选出的10对SSR适宜引物,4个新品系及亲本共扩增出稳定而清晰的SSR条带142个,多态性位点百分率达81.69%;建立了能区分出4个优异新品系及其亲本的SSR指纹图;4个杂种新品系及其双亲的GD值变幅为0.229 8~0.650 8。以GD值0.34为基准,7个马铃薯材料分为3类:第1类为新品系A 2、A 10和H-027,第2类为新品系B 1、B 12和陇薯7号,陇薯6号单独为1类。In order to determine the genetic differences of new strains of potato in DNA level, SSR molecular marker technology was used for polymorphic analysis of four excellent new strains (A 2, A 10, B 1 and B 12) from the F1 hybrids of two hybrid combinations H-027 X Longshu No.6 and H-027 XLongshu No.7.Using 10 SSR suitable primers to amplify genome DNA of four new srains and its' parents, a total of 142 clear and stable SSR polymorphic bands were detected, and the polymorphic rate was 81.69%.A SSR fingerprint map was established, which could distinguish four excellent new strains and their parents.The genetic distance (GD) of test plants were from 0.229 8 to 0.650 8.This threshold 0.34 was adopted as GD value, seven tested plants were classified into three categories: the new strain A 2,A 10 and H-027 as a category;B 1,B 12 and Longshu No.7 as a category;and Longshu No.6 as separate category.
