摘  要:(1)目的观察丁苯酞(dl-3-n-butylphthalidle,NBP)对阿尔茨海默病(Alzheimer disease,AD)模型大鼠学习记忆能力及海马CA1区超微结构的影响,探讨NBP对AD模型大鼠的脑保护作用。(2)方法雄性SD大鼠18只,分为假手术组、AD模型组、NBP治疗组。AD组和Treatment组采用β-淀粉样蛋白1-42(beta-amyloid people 1-42,Aβ1-42)海马立体定向微量注射的方法建立模型,Sham组采用同样的方法注射生理盐水,4周后Morris水迷宫实验比较3组大鼠逃避潜伏期及穿越平台次数。而后Treatment组给予NBP溶液进行灌胃;Sham及AD组采用同等剂量食用油灌胃,2次/d。2w后进行Morris水迷宫实验检测各组大鼠学习记忆能力,电镜下观察各组大鼠海马CA1区超微结构的改变。(3)结果 4周时与Sham组比较,AD组和Treatment组逃避潜伏期明显延长、穿越平台次数明显减少(P<0.05);AD组与Treatment组比较无明显差异(P>0.05)。6周时与Sham组比较,AD组和Treatment组逃避潜伏期显著延长、穿越原平台位置次数减少(P<0.05);与AD组相比较,Treatment组逃避潜伏期明显缩短、穿越平台位置次数明显增多(P<0.05),电镜下可见Treatment组神经元损伤较AD组有所减轻。(4)结论 NBP能够改善AD模型大鼠的学习记忆能力,减轻海马CA1区神经元的受损程度。Objective To explore neural protection of dl-3-n-butylphthalidle(NBP) in Alzheimer's disease(AD)model rats by observing the effect on learning and memory ability and the ultrastrure in hippoeampal CA1 region.Methods 18 male SD rats were randomly divided into three groups:sham operation group,AD model group,NBP treatment group.Animal models of AD group and the Treatment group were established with 5ug beta-amyloid 1-42 by Stereotaetic mieroinjection of bilateral hippoeampus,and the Sham group with equel sailine. After 4 weeks, Morris water maze experiment were tested,compared by escape latent period and numbers of cross platform of rats in three groups. Then the treatment group were given NBP by intragastric administration, and the Sham group and the AD group treated oniy by the same dose of cooking oil,2 times a day.Another 2 weeks later,Mor- ris water maze experiment were tested again to detect the ability of learning and memory,the change of ultrastrure of rats in hippocampal CA1 region by electron microscope were observed.Results In the forth week,Morris water maze experiment showed that the escape latent period of rats in the AD group and the Treatment group prolonged significantly, the number of cross platform in the two groups reduced(P〈0.05).Compared with the AD group,there was no statistically significance in the Treatment group(P〉0.05).At 6th week, Morris water maze experiment showed that the escape latent period of rats in the AD group and the Treatment group prolonged significantly, the number of cross platform in the two groups reduced(P〈0.05).Compared with the AD group,escape latent peroid of rats in the Treatment group shorten significantly,whereas the number of cross platform of rats in the Treatment group exceeded obviously(P〈0.05).Compared with the AD group,the electron mi- croscope showed that in hippocampal CA1 region nerve damage of the Treatment group relieved.Con- clusion NBP can improve the ability of learning and memory,reduce the nerve cell damage in hi
