摘  要:目的:探讨CD25基因多态性与云南汉族人群肺癌发生发展的相关性。方法:选取云南地区汉族人群肺癌Ⅰ+Ⅱ期患者151例,肺癌Ⅲ+Ⅳ期患者316例,健康体检人群384例,采用Taq Man探针基因分型方法对CD25基因中2个多态性位点rs7072793(C>T)和rs3118470(C>T)进行基因分型,研究其等位基因、基因型及所构建的单倍型在肺癌Ⅰ+Ⅱ期患者、肺癌Ⅲ+Ⅳ期患者和健康对照人群中频率分布的差异。结果:rs7072793和rs3118470位点等位基因在各组中的分布频率比较,差异无统计学意义(P>0.05),两个位点等位基因可能与肺癌的发生发展无关;遗传模式分析的结果经年龄校正后,rs7072793位点采用最优遗传模式Recessive分析发现,基因型CC-CT和TT在Ⅰ+Ⅱ期组和Ⅲ+Ⅳ期组中的分布频率比较,差异有统计学意义(P=0.004);rs3118470位点采用最优遗传模式Dominant分析发现,基因型CT-CC和TT基因型在Ⅰ+Ⅱ期组和Ⅲ+Ⅳ期组中分布频率比较,差异有统计学意义(P=0.019);rs7072793位点基因型CC-CT可能是肺癌发生的保护性因素(OR=0.45,95%CI:0.25~0.80),rs3118470位点基因型TT可能是肺癌发生的风险因素(OR=1.79,95%CI:1.09~2.95)。结论:CD25基因中多态性位点rs7072793位点基因型CC-CT可能是肺癌发生的保护性因素,rs3118470位点基因型TT可能是肺癌发生的风险因素。Objective: To evaluate the association of the single nucleotide polymorphisms(SNPs) in CD25 gene with the occurrence and development of lung cancer in Yunnan Han population. Methods: A total of 155 patients in pathological stages Ⅰ+Ⅱ, 316 patients in pathological stages Ⅲ+Ⅳ and 384 healthy individuals were recruited in this study. TaqMan probe genotyping was adopted to conduct genotyping in the 2 polymorphic loci rs7072793 ( C 〉 T) and rs3118470 ( C 〉 T), and then the fre- quency distribution differences of the alleles. Results: The results showed that there was no significant difference of allelic frequencies in the 2 polymorphic loci rs7072793 ( C 〉 T) and rs3118470 (C 〉T) between I + lI, Ⅲ+Ⅳ and control groups, suggesting that the two allele were not related to the occurrence and development of lung cancer. After the genetic model analysis results was adjusted with age, it was found that after optimal model Recessive analysis on rs7072793 loci, there were statistically significant differences of frequency distribution in genotype CC-CT and IT be- tween pathological stagesⅠ+Ⅱ and pathological tages Ⅲ+Ⅳ group( P =0. 019) , suggesting that rs7072793 genotype CC-CT might be a protective factor of lung cancer occurrence (OR = 0.45, 95% CI: O. 25 -0.80) while rs3118470 genotype TT might be a risk factor of lung cancer occurrence( OR = 1.79,95% CI: 1.09 - 2.95 ). Conclusion: in CD25 gene, the polymorphic loci rs7072793 geuo- type CC-CT may be a protective factor of lung cancer occurrence, but rs3118470 genotype IT may be a risk factor of lung cancer occurrence.
