摘 要:目的筛选丙酸睾丸酮诱导去势大鼠前列腺增生模型的成模最佳时间点,并在此过程中观察前列腺组织的动态病理变化。方法 48只成年雄性SD大鼠随机分成6组,每组8只:对照组、去势组、实验组根据去势后皮下注射丙酸睾丸酮后取材时间分为7d组、14d组、21d组、28d组;测量各组大鼠体重、前列腺体积、湿重及前列腺指数;采用HE染色法观察前列腺组织病理学改变;Western blot法检测大鼠前列腺组织IL-6蛋白的表达情况。结果大鼠去势后前列腺组织萎缩。与对照组及7d组比较,14d组、21d及28d组大鼠前列腺体积、前列腺湿重、前列腺指数均增加(P<0.05)。且21d组与28d组前列腺体积较14d组增大(P<0.05),但21d组与28d组间各数值差异无统计学意义(P<0.05)。14d组大鼠前列腺上皮细胞和间质组织出现增生改变,21d组和28d组大鼠前列腺上皮细胞和间质组织明显增生,且出现炎性细胞浸润现象,28d组更为明显。与对照组比较,28d组大鼠前列腺组织IL-6的表达升高(P<0.05)。结论去势后皮下注射丙酸睾丸酮21天就可成功构建大鼠前列腺增生模型,且在此过程中伴随慢性前列腺炎症的发生。Objective To screen the optimal time point changes in the prostate tissue in the process. Methods for modeling and to observe the dynamic pathological 48 adult male SD rats were randomly divided into six groups: control group, Castration group, 7d(days ) model group, 14d model group, 21d model group, 28d model group, 8 rats for each group. Body weight, prostate volume, prostate wet weight and prostate index ( prostate wet weight/body weight) were measured in rats of all groups. HE staining was performed to observe the histopathological changes of the prostate. IL-6 protein expression were detected by western blotting. Compared with control and 7d group, rat prostate volume, prostate wet weight and prostatic index were significantly increased in 14d, 21d and 28d group (P〈0.05). Compared with 14d group, prostate volume was increased in 21d and 28d group. Compared with 21d group, there was no statistically significant difference among prostate volume, prostate wet weight and prostatic index in 28d rat group. HE staining indicated proliferation of the epithelial cells and stromal cells were visible in 14d group while hyperplasia and inflammatory cell infiltration developed in the epithelial and stromal tissue in 21d and 28d group. Compared with control group, protein expression of IL-6 in the rat prostate tissue was significantly increased in 28d group (P〈0.05). Conclusion After subcutaneous injection of testosterone propionate in castration rats, the needed models can be established at the 21st days. Moreover, chronic inflammation of the prostate will develop as complication during modeling.
