摘 要:目的探讨替格瑞洛含药血清对人胃黏膜上皮细胞株GES-1的损伤作用。方法分离1例服用替格瑞洛(180mg/d)后2h的患者血清,并配制成不同浓度(5%、10%、15%、20%)的含药血清,同时取该患者服用替格瑞洛前的血清作为对照,培养GES-1细胞24、48、72h。采用MTT法检测细胞增殖,倒置相差显微镜观察各浓度组含替格瑞洛血清培养GES-1细胞后细胞形态学变化,流式细胞仪检测各浓度组细胞凋亡情况。结果MTr显示替格瑞洛含药血清对GES-1细胞的损伤随着浓度增加而增强,呈浓度依赖性(P〈0.05),而无时间依赖性(P〉0.05)。光镜下可见含药血清组的贴壁细胞数量减少,细胞变圆、悬浮,部分细胞核浓缩,细胞损伤随着含药血清浓度增加而加重。流式细胞术显示,替格瑞洛含药血清组细胞凋亡率增大,且随浓度增加而增强。结论替格瑞洛含药血清对人胃黏膜上皮细胞有损伤作用.Objective To investigate the effects of ticagrelor on human gastric mucosal epithelial GES - 1 cells. Methods Serum was collected from a patient taking 180 mg/d ticagrelor 2 h after treatment, and various concentrations (5% , 10% , 15% , and 20% ) of ticagrelor in serum were prepared. Meanwhile, the serum separated from the same pa- tient before ticagrelor treatment was taken as a control. GES - 1 cells were incubated with the ticagrelor - containing ser- um for 24, 48 and 72 h. The proliferation rate of GES - 1 cells was examined using MTF assay. The morphological chan- ges of GES - 1 cells treated with different concentrations of ticagrelor for 24 h were observed under an inverted phase con- trast microscope. Flow cytometry was used to detect cell apoptosis after GES - 1 cells incubated with the above serum. Results According to MTI" assay, ticagrelor inhibited the proliferation of GES - 1 cells in a concentration - dependent manner (P 〈 0.05) , rather than in a time - dependent manner (P 〉 0.05 ). Compared with the control group, the ti- cagrelor - containing serum group showed a less number of adherent cells, where the cells became round and suspended, some cells presented condensed nucleus and cellular damages gradually worsen as the concentration of ticagrelor was in- creased. According to flow cytometry, the apoptosis rate of GES - 1 cells was increased in a dose dependent manner 'after treated with ticagrelor. Conclusion Ticagrelor can result in damages in human gastric mucosal epithelial GES - 1 cells.
