摘 要:目的研究番茄红素对丙烯腈(ACN)染毒大鼠睾丸的保护性机制。方法将72只雄性SD大鼠随机分成6组,分别为对照组(Control)、染毒组(ACN)、泰特(TAD)组(ACN+TAD),低剂量(10mg/kg)、中剂量(20mg/kg)和高剂量(30mg/kg)番茄红素组。对各组睾丸组织脂质过氧化产物(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)水平进行检测,对睾丸组织病理切片HE染色及NF-κB p65免疫组化染色。结果与对照组比较,染毒组大鼠MDA含量升高,而SOD和GSH活力水平均下降,组织形态明显损伤,NF-κB p65表达升高,差异有统计学意义(P<0.05);番茄红素处理以后,特别是中、高剂量番茄红素处理以后,MDA含量明显降低,而SOD、GSH活力水平上升,同时组织形态损伤程度减轻,NF-κB p65的表达降低,差异有统计学意义(P<0.05)。结论番茄红素对ACN染毒所致大鼠睾丸损伤有一定的保护作用,且这种保护作用可能与减少氧化应激和降低NF-κB的表达有关。Objective To study the effect of lycopene on testis in acrylonitrile infected rat and its possible mechanism. Methods 72 male SD rats were randomly divided into 6 groups, namely the control group (control), infected group(ACN), TAD group (ACN +TAD), low dose group, middle dose and high dose of lycopene group respectively. The MDA, SOD, GSH-px levels of testicular tissue were tested in each group. Pathological section and HE staining for testicular tissue, and immunohistochemical staining on NF-κB p65 were performed. Results Compared with the control group, the MDA level increased and SOD, GSH level decreased in infected group, as well as obvious morphological damage was observed, the NF-κB p65 expression increased, these differences were statistical significance. After treated by lycopene, especially by moderate dose and high dose, MDA level decreased significantly, while SOD and GSH levels rose, tissue injury relieved, NF-eB p65 expression decreased. A statistically significant difference was found. Conclusions Lycopene has a protective effect to rat testicular damage caused by enenitrile, and this protective effect may be related to reduce oxidative stress and to reduce the expression of the NF-κB.
