摘  要:目的本文试图在羊急性心梗模型上建立miRNA表达谱,为LVAD治疗急性心梗提供理论依据。方法成年小尾寒羊经结扎冠状动脉制作心梗模型,心脏不停跳植入FW-2型轴流泵,连续卸负荷循环辅助3天。取出的心脏用组织染色法检测。高通量测序技术建立miRNA表达谱。结果对照组动物实验共进行7只,其中3只造模成功。LVAD组动物实验共有5只,其中3只成功。miRNA测序结果共发现已知miRNA成熟体152个,新miRNA成熟体1582个。卸负荷组与对照组相比发现差异表达,其中oar-miR-19b(上调)和oar-miR-26a(上调)得到RT-PCR验证,与芯片结果相符。GO和KEGG分析发现这些mi R参与调节的功能及信号通路与心血管疾病密切相关。结论本实验利用FW-2型轴流泵成功地对急性心梗的羊进行了3天的循环辅助,证实LVAD可有效减轻心梗程度。成功地建立了急性心梗和LVAD干预的miRNA表达谱,并进行了验证,结果表明该表达谱可用于探索未知的miRNA,并研究它们参与急性心梗反应的调控机理。Objective To establish a miRNA expression profile in the acute myocardial infarction model of sheep, and to provide theoretical basis for the treatment of acute myocardial infarction with LVAD. Methods The adult small - tail Hart sheep was treated with ligation of the coronary artery. The heart was anchored into the FW - 2 axial - flow pump, and the continuous unloading cycle was assisted for 3 days. The removed heart was stained with tissue staining. High - throughput sequencing technology to establish miRNA expression profiles. Results There were 7 animals in the control group, 3 of which were successfully modeled. LVAD group of animals, a total of five experiments, of which three were successful. MiRNA sequencing results were found in 152 miRNA matures, 1582 miRNA matures. The oar-miR-19b (up-regulated) and oar-miR-26a (up-regulated) were identified by RT-PCR and were consistent with the results of the chip, compared with the control group. GO and KEGG analysis revealed that these miRs were involved in the regulation of function and signaling pathways associated with cardiovascular disease. Conclusions In this study, FW-2 axial pump was successfully used to support the acute myocardial infarction for 3 days. It was confirmed that LVAD could effectively reduce the degree of myocardial infarction. The miRNA expression profiles of acute myocardial infarction and LVAD intervention were successfully established and verified. The results showed that the expression profile could be used to explore the unknown miRNAs and study their regulation mechanism of acute myocardial infarction.
