摘  要:目的:探讨血清反应因子(serum response factor,SRF)-早期即刻基因(immediate early gene,IEG)在慢性束缚应激模型大鼠内脏高敏感形成中的作用。方法:将18只大鼠随机分成不给予束缚对照组、束缚模型组1和束缚模型组2(无结肠直肠扩张),每组6只。用束缚前后结直肠扩张引起的腹壁回撤反射评分(AWR评分)3分时最小扩张球囊压力(即痛阈)来评价大鼠内脏敏感性的变化,采用Western blot法检测各组SRF和IEG(c-fos和Arc)及SRF-IEG下游的突触变化相关蛋白的表达情况。结果:(1)模型组1束缚后痛阈显著低于束缚前和对照组(P<0.05)。(2)模型组1和模型组2的SRF、c-fos和Arc表达比对照组显著增强(P<0.05),而2个模型组间差异无统计学意义(P>0.05)。(3)2个模型组的突触素、PSD-95、NMDA-R1、p-Ca MKⅡ4个因子表达比对照组显著增强(P<0.05),而2个模型组间差异无统计学意义(P>0.05)。结论:SRF-IEG通路在慢性束缚应激所致大鼠内脏高敏感形成中发挥一定作用。Objective: To investigate the role of SRF-IEG pathway in rats with visceral hypersensitivity caused by chronic restraint stress. Methods: A total of 18 rats were randomly allocated to three groups equally, including control group, restraint group 1 and restraint group 2 without colorectal distension (CRD) . Control group and restraint group 1 received CRD. The minimum pressure was recorded as pain threshold when the AWR score was three. The formation of visceral hypersensitivity was evaluated by pain threshold changes. The expression of SRF, IEG ( including Arc and c-fos), synaptophysin, PSD-95, NMDAR1 and p- CaMKII in dorsal root ganglion (DRG) of rats were detected by Western blot. Results: (1) The pain threshold in restraint group 1 was markedly lower than that in control group and before restrtraimt (P〈0.05) . (2) The expressions of SRF, Arc and c-fos were significantly higher in two restraint groups than those in control group (P〈O.05) . No significant difference was found between the restraint group land the restraint group 2. (3) The expressions of synaptophysin, PSD-95, NMDAR1 and p-CaMKII were significamtly higher in two restraint groups than those in control group (P〈0.05) . No significant difference was found between the restraint group 1 and the restraint group 2. Conclusion: The SRF-IEG pathway possibly plays roles in the formation of visceral hypersensitivity caused by significamtly chronic restraint stress.
