摘 要:目的探讨Raf-1、MEK-2、ERK-1在胃癌组织中的表达形式及其意义,并分析此种信号途径与胃癌的临床病理特征之间是否存在联系,并为胃癌抗EGFR靶向治疗的分子标志物筛选提供依据。方法通过免疫组化中SP法检测60例胃癌患者标本胃癌组织和正常的胃组织中Raf-1、MEK-2、ERK-1的表达水平并进行比对。结果经统计60例胃癌组织当中Raf-1、MEK-2、ERK-1的表达率分别为88.33%(53/60)、85.00%(51/60)、78.33%(47/60)。而正常的胃组织当中Raf-1、MEK-2、ERK-1均未见到阳性表达。被胃癌细胞转移的淋巴结组织中Raf-1、MEK-2、ERK-1的阳性表达率更为显著,分别高达95.12%(39/41),90.24%(37/41),87.80%(36/41)。未见癌细胞转移的淋巴结中Raf-1、MEK-2、ERK-1均无表达。实验证实Raf-1、MEK-2、ERK-1的表达水平与胃癌的临床特征中的肿瘤分化程度、浸润深度、淋巴结转移以及TNM分期有明显相关性(P<0.05)。结论 (1)人体胃癌组织当中存在Raf/MEK/ERK信号通路的激活;(2)人体胃癌组织中Raf/MEK/ERK信号通路的活化程度与胃癌组织的临床病理特征相关,此信号通路活化程度越高的胃癌组织,其分化的程度越低,TNM分期越高。三者联合检测可为研究胃癌的生物学行为乃至对靶向治疗胃癌提供十分重要的参考依据及指标。Objective To investigate the expression pattern and significance of Raf-1, MEK-2 and ERK-1 in gastric carcinoma and analyze whether there is a link between this signaling pathway and the clinicopathological features of gastric cancer, and to provide the basis for the screening of molecular markers for gastric cancer .anti-EGFR targeted therapy. Methods The expression levels of Raf-1, MEK-2 and ERK-1 in gastric cancer tissues and normal gastric tissueswere detected by SP immunohistochemical method and compared in 60 patientswith gastric cancer. Results The expression rates of Raf-1, MEK-2 and ERK-lwere 68.33% (53/60), 85.00% (51/60) and 78.33% (47/60) respectively in 60 cases of gastric cancer. However, the positive expressions of Raf-1, MEK-2 and ERK-1 were not observed in normal gastric tissues. The positive expression rates of Raf-1, MEK-2 and ERK-1 in lymph node metastasis of gastric cancer cells were 95.12%(39/41), 90.24%(37/41), 87.80%(36/41), which were significantly higher. Raf-1, MEK-2 and ERK-1 were not expressed in lymph nodes without lymph node metastasis. The expression levels of Raf-1, MEK-2 and ERK-1 were significantly correlated with the degree of tumor differentiation, depth of invasion,lymph node metastasis and TNM staging in the clinical features of gastric cancer(P〈0.05). Conclusion (1) The activation of Raf/MEK/ERK sig- naling pathway is existed in human gastric cancer tissues; (2)The activation degree of Ratf/MEK/ERK signaling pathway in human gastriecancer tissue is related to elinicopathologieal features of gastric cancer tissue. The higher the degree of activation of this signal pathway in gastric cancer tissues, the lower the degree of differentiation, and the higher the TNM staging. The combined detection of the three can provide a very important reference and indicator for the study of the biological behavior and the targeted treatment of gastric cancer.
