摘  要:目的检测结直肠癌、结直肠腺瘤及正常结直肠黏膜中Pokemon的表达,探讨结直肠腺瘤恶变过程中Pokemon的作用及价值。方法收集结直肠癌60例、结直肠腺瘤80例、结直肠正常组织40例。采用免疫组织化学技术及荧光定量聚合酶链反应(PCR)技术检测Pokemon在结直肠癌、结直肠腺瘤及正常结直肠黏膜中的表达。结果免疫组织化学技术结果显示正常结直肠黏膜、结直肠腺瘤、结直肠癌中Pokemon的阳性表达率分别为25.0%、51.2%、80.0%,三者之间差异有统计学意义(P=0.000)。结直肠管状腺瘤、结直肠混合性腺瘤、结直肠绒毛状腺瘤中Pokemon的阳性表达率分别为33.3%、53.3%、75.0%,差异有统计学意义(P=0.015)。Pokemon的表达与淋巴结转移(P=0.040)、肿瘤的分化程度(P=0.004)、Dukes分期(P=0.014)有关,而与年龄(P=0.947)、性别(P=0.795)无明显相关。荧光定量PCR显示Pokemon特异性扩增,Pokemon在正常结直肠黏膜、结直肠管状腺瘤、结直肠混合性腺瘤、结直肠绒毛状腺瘤、结直肠癌组织中的表达分别为0.981±0.189、2.024±0.226、3.357±0.262、5.006±0.281、6.450±0.216,F=432.884,P=0.012。结论Pokemon参与了结直肠腺瘤、结直肠癌的发生与发展。Objective To detect the expression levels of Pokemon in colorectal carcinoma, colorectal adenomas and normal colorectal mucosa, and analyze the relationships among different histological types of colorectal adenomas and colorectal carcinoma. Methods Sixty cases of colorectal carcinoma tissues, 80 cases of colorectal adenomas, and 40 cases of normal colorectal mucosa tissues were collected. Immunohistochemistry and fluorescence quantitative polymerase chain reaction (PCR) were used to detect the expression of Pokemon in the tissues. Results The immunohistochemistry revealed that the positive ex- pression rate of Pokemon protein in normal colorectal mucosa, colorectal adenoma and colorectal carcinoma tissues was 25.0%, 51.2% and 80. 0%, respectively, with significant differences among them ( P = 0. 000). The positive expression rate of Pokemon in tubular adenoma, mixed pattern of adenoma and vil- lous adenoma was 33.3%, 53.3% and 75. 0% , respectively, also with significant differences among them (P =0. 015 ). There was a significant correlation between the raised expression of Pokemon and unfavor- able variables, including nodal metastasis ( P = 0. 040), Dukes stage ( P = 0. 014), and differentiation de- gree (P = O. 004). There was no obvious difference in age ( P = 0. 947 ) and sex ( P = 0. 795 ). Fluores- cence quantitative PCR showed characterized amplification about Pokemon. The expression levels of Poke- mon mRNA in normal colorectal mucosa, tubular adenoma, mixed pattern of adenoma, villous adenoma and colorectal carcinoma were 0. 981 ± 0. 189, 2. 024 ± 0. 226, 3. 357 ± 0. 262, 5. 006 ± 0. 281, and 6. 450 ± 0. 216 respectively,F = 432. 884,P = 0. 012. Conclusion Pokemon participates in the occurrence and development of colorectal adenoma and colorectal carcinoma.
