摘　　要：目的探讨泛素样含PHD和环指域1(UHRF1)基因在人原发性肝细胞肝癌中的表达及其与临床病理特征的关系。方法采用半定量逆转录聚合酶链式反应(RTPCR)、免疫组织化学法检测63例肝癌组织、癌旁组织及10例正常肝组织中UHRF1 mRNA及蛋白的表达情况,分析UHRF1蛋白表达与肝癌患者临床病理参数及预后的相关性。结果肝癌组织中UHRF1 mRNA表达水平明显高于癌旁组织和正常肝组织(P均<0.05);肝癌组织中UHRF1蛋白阳性表达率为85.71%(54/63),而UHRF1在癌旁组织几乎不表达。UHRF1的表达与肿瘤分化、大小、TNM分期及是否侵袭周围组织密切相关(P均<0.05),与患者预后呈负相关(P<0.05)。结论 UHRF1在肝癌组织中高表达,和多项临床病理因素具有明显关联,其在肝癌早期诊断、预后判断及靶向治疗中具有潜在的临床应用价值。Objective It is to investigate the expression of UHRF1 in primary hepatocellular carcinoma （HCC） and the correlation with clinicopathological features. Methods The expression of UHRF1 was detected in 63 surgically resected specimens, corresponding adjacent and 10 normal liver tissues of HCC by reverse transcription-polymerase chain reaction （RT- PCR） and immunohistochemistry. The relationship of UHRF1 with the clinicopathological characteristics of HCC were ana- lyzed. Results The UHRF1 mRNA level was significantly increased in HCC tissues compared with adjacent and normal liver tissues （all P 〈 O. 05 ）. There were 85.71% （54/63） samples of positive for UHRF1 in these tumor specimens, however was almost negative in adjacent tissues. The abnormal expression of UHRF1 was correlated with the differentiation of tumor, size, TNM stage and invasion （ all P 〈 0.05 ）, meanwhile was negatively correlated with postoperative prognosis （P 〈 0.05 ）. Con- clusion UHRF1 is overexpressed in the tissues of HCC, and UHRF1 expression is highly correlated with several clinical pa- rameters. Therefore, UHRF1 plays important roles in early prediction, diagnosis and survival of HCC. Blockade of UHRF1 signaling pathway may have a therapeutic role in HCC treatment.
