摘  要:目的:探讨神经胶质瘤组织中微小RNA-132(miR-132)的表达及临床意义。方法:采用实时荧光定量聚合酶链反应(qRT-PCR)方法检测新鲜神经胶质瘤组织标本135例及正常脑组织标本53例中miR-132的表达,分析其与神经胶质瘤的临床病理特征关系、诊断价值及与预后的关系。结果:miR-132在神经胶质瘤组织中的表达显著高于正常组织(P<0.05);miR-132在神经胶质瘤组织高表达和低表达组在肿瘤切除程度、WHO分级、KPS评分方面差异均具有统计学意义(均P<0.05);ROC曲线分析AUC=0.856(P<0.001),灵敏性和特异性分别是52.7%和86.6%,Youden指数为0.465;miR-132高表达和低表达组在总生存时间及无进展生存时间上的差异具有统计学意义(均P<0.05)。结论:miR-132在神经胶质瘤中高表达的,可能可作为神经胶质瘤的新型生物标记物和诊断靶标。Objective: To investigate the expression and clinical significance of microRNA (miR-132) in pa- tients with glioma. Methods: One hundred thirty-five cases with glioma (glioma group) and 53 cases of normal brain tissues (control group) were collected. Real-time quantitative polymerase chain reaction (qRT-PCR) was used to detect the miR-132 expression in 2 groups. The clinic pathological characteristics ofmiR-132 expression were further analyzed. A receiver operating characteristic (ROC) curve was constructed to determine the diagnos- tic value of miR-132. Results: miR-132 expression was significantly up-regulated in the glioma group com- pared with that in the normal group (P〈0.05). There was significant differences between KPS score, extent of re- section and WHO grade in terms of miR-132 expression (P〈0.05). The area under the ROC curve was 0.856 (P〈0.001), the sensitivity was 52.7%, the specificity was 86.6% and the index of Youden was 0.465. The overall survival time and progression free survival in miR-132 high expression and low expression sub-groups were sta- tistically significant (P〈0.01). Conclusion: miR-132 may be involved in carcinogenesis of glioma, which may be a new biomarker for diagnosis and treatment.
