摘 要:试验首先采用高通量测序法检测常温保存1、3、5 d关中黑猪精液细菌的种类及所占比例,而后采用16S rDNA法初步鉴定精液中分离的部分细菌,并筛选出精液中常见细菌作为ε-聚赖氨酸体外抑菌试验菌种,最后探究0、0.01、0.02、0.04、0.08、0.16、0.32 g/L的ε-聚赖氨酸对精液中常见细菌的抑制作用。结果表明,在常温保存过程中革兰氏阴性菌为优势菌群,且0.04 g/L的ε-聚赖氨酸能有效抑制大肠杆菌、表皮葡萄球菌和鲁菲不动杆菌的生长。因此,在常温保存过程中添加ε-聚赖氨酸能有效控制精液中常见细菌的生长。The experiment was conducted to explore the inhibitory effect of e-polylysine on common bacteria in semen and to provide new ideas for reducing the use of antibiotics in dilutions. First, the high-throughput sequencing method was used to detect the species and proportion of semen bacteria in the black boar at room temperature for 1, 3, 5 days. Then, 16S rDNA method was used to identify some bacteria isolated from semen and the bacteria ε-polylysine in vitro, and finally explored 0 g/L, 0.01 g/L, 0.02 g/L, 0.04 g/L, 0.08 g/L, 0.16 g/L, 0.32 g/L ε-polylysine inhibits common bacteria in semen. The results showed that Gram-negative bacteria were the dominant flora during the preservation of normal temperature, and 0.04 g/L ε-polylysine could inhibit the growth of Escherichia coli, Staphylococcus epidermidis and Acinetobacter lwoffi. Therefore, the addition of ε-polylysine during the preservation of the room temperature can effectively control the growth of common bacteria in semen.
