摘 要:结合形态特征和分子生物学方法,将一株分离自川贝母鳞茎的内生真菌CBY11鉴定为Fusarium tricinctum。分别研究其发酵液石油醚提取物(PEE)、乙酸乙酯提取物(EAE)、正丁醇提取物(BAE)和乙醇提取物(ETE)的DPPH自由基清除活性、总抗氧化活性(以ABTS法测定)和总还原力(FRAP法测定),以及其总多酚含量(TPC)、总黄酮含量(TFC)和总皂苷(TSC)含量。结果发现EAE、BAE和ETE均具有较强的DPPH自由基清除活性(IC50分别为1.95mg/m L、1.19mg/m L、1.64mg/m L)和总抗氧化活性(IC50分别为0.43mg/m L、0.58mg/m L、0.43mg/m L),但总还原能力相对较弱;PEE的抗氧化活性整体较低。此外,EAE、BAE和ETE的TPC、TFC及TSC显著高于PEE(P<0.05)。可见TPC、TFC及TSC与抗氧化活性呈现出一定的正相关性。HPLC分析表明EAE含有多种具有较强抗氧化活性的酚酸等物质,如芹菜素。GC/MS分析表明PEE中含有少量抗氧化酚类物质,最主要成分是烷烯烃类。本研究表明川贝母内生真菌CBY11具有产生丰富抗氧化活性成分的能力,具有多种利用价值。An endop biological methods hytic fu and mo ngus isolated from Fritillaria cirrhosa was identified as Fusarium tricinctum by combining molecular r assay), the reduction power phological characteristics. The DPPH (FRAP assay), total phenolic content radical scavenging activities, total antioxidant capacities (ABTS (TPC), total flavonoid content (TFC) and total saponin content (TSC) of the petroleum ether extract (PEE), ethyl acetate extract (EAE), n-butyl alcohol extract (BAE) and ethanol extract (ETE) of the strain CBY11 of the fungus were determined. The results indicated that EAE, BAE and ETE exhibited higher DPPH radica scavenging activities (IC_50 values 1.95mg/mL, 1.19mg/mL and 1.64mg/mL, respectively), higher total antioxidant capacities (IC_50 values 0.43mg/mL, 0.58mg/mL and 0.43mg/mL, respectively) and relatively lower reduction power. The antioxidant activities of PEE were totally lower. TFC, TPC and TSC of EAE, BAE and ETE were significantly higher than those of PEE (P〈0.05). There were positive correlations between TFC, TPC and TSC and the antioxidant activities to a certain degree. HPLC analysis indicated that EAE possessed multiple phenol acid compounds with higher antioxidant activities, such as apigenin. GC/MS analysis revealed that PEE had a few phenol acid compounds with antioxidant activities, mainly hydrocarbon compounds. The present study reveals that endophytic fungus Fusarium tricinctum CBY11 has ability producing plentiful bioactive compounds with antioxidant properties, being possessed of multiple utilization values.
