摘  要:目的:探讨乳腺术后骨转方对骨癌痛大鼠疼痛行为及脊髓星形胶质细胞神经胶质纤维酸性蛋白(GFAP)表达的影响。方法:SD大鼠54只,按体重随机分成6组,每组9只,于大鼠左后肢注射Walker-256细胞建立骨癌痛模型,观察各组大鼠造模后热痛敏阈值的变化,28 d后取大鼠脊髓腰膨大处,检测GFAP mRNA和蛋白、SP和CGRP蛋白的表达情况。结果:(1)乳腺术后骨转方各剂量组在造模后第21天的PWTL高于模型组(P<0.05),至造模后28 d,乳腺术后方高剂量+唑来膦酸组PWTL仍高于Model组(P<0.05)。(2)乳腺术后骨转方各剂量组可以抑制造模后28 d脊髓中GFAP mRNA和蛋白的表达,与Model组比较有统计学意义(P<0.05)。Z+H组和H组的SP蛋白表达明显低于Model组,统计学上差异显著(P<0.05);各给药组可以抑制脊髓中CGRP蛋白的表达(P<0.01)。其中Z+H组作用优于其他各给药组,统计学上差异显著(P<0.05)。结论:乳腺术后骨转方具有镇痛作用,抑制脊髓水平星形胶质细胞GFAP、SP、CGRP的表达可能是其作用机制之一。Objective : To investigate the effect of postoperative breast Guzhuan Decoction on pain behavior and expression of glial fibrillary acidic protein ( GFAP ) of spinal cord astrocytes activation in the rats tibial cancer pain model. Methods: 54 SD rats were randomly divided into 6 groups according to the weight, each group had 9 rats. Inject Walker 256 mammary gland carcinoma cells into the tibia medullary cavity via intercondylar eminence to prepare the bone cancer pain model. Observed the PWTL changes in rats after model made, took the enlargement of lumbar spine on the 28th day to detection the protein and mRNA expression on GFAP, protein expression on SP and CGRP. Results : ( 1 ) The PWTL of each dosage group of postoperative breast Guzhuan Decoction was longer than model group on the 21th day ( P〈0.05 ), the PWTL of Z+H group was still higher than model group on the 28th day after model made ( P〈0.05 ). ( 2 ) Each dosage group of postoperative breast Guzhuan Decoction could restrain the protein and mRNA expression on GFAP, there was a statistical significance compare with model group ( P〈0.05 ). The protein expression of SP was significantly lower than model group ( P〈0.05 ). Each dosage group of postoperative breast Guzhuan Decoction could restrain the protein and expression on CGRP (P〈0.01), the Z+H group was significantly better than other group (P〈0.05). Conclusion: Postoperative breast Guzhuan Decoction had analgesic effect, inhibition GFAP, SP, CGRP expression of spinal cord astrocytes might be one of the mechanisms.
