摘  要:目的:研究自拟醒脑除颤方对6-羟基多巴诱导帕金森病模型大鼠的增效减毒作用。方法:将SD大鼠的大脑定位于内侧前脑束(Medial forebrain bundle,MFB)部位再注入6-羟基多巴(24μg)制成PD模型大鼠。再把成功的PD大鼠随机分为5组,每组10只,雌雄各半,即模型组、美多巴组(75 mg/kg)、左旋多巴组(60 mg/kg)、β-细辛醚组(15 mg/kg)、自拟醒脑除颤方组(左旋多巴60 mg/kg+β-细辛醚15 mg/kg)。给药组按上述剂量灌胃给药,1 d 2次,给药30 d。给药结束观察各组5 min内的AIM评分(口面部、轴性和前肢部位),计算肝肾系数和HE染色观察纹状体的组织病理形态。结果:与假手术组比较,模型组的口面部、轴性和前肢的AIM评分都显著增加(P<0.01),肝肾系数无显著差异(P>0.05),纹状体中神经细胞较少,残留神经元核固缩,部分细胞胞浆明显肿胀,这说明了PD大鼠造模成功和造模剂6-羟基多巴对模型大鼠无肝肾毒性。与模型组比较,各给药组的AIM评分都显著减少(P<0.01),给药治疗后纹状体神经细胞数略有增多,结构基本正常,无明显炎性浸润变性及坏死现象,其中左旋多巴组的肾脏系数显著增加(P<0.05)。与左旋多巴组比较,美多巴组和自拟醒脑除颤方组的口面部、轴性和前肢的AIM评分及右肾系数都显著减少(P<0.05)。结论:自拟醒脑除颤方对6-羟基多巴诱导帕金森病模型大鼠具有一定增效减毒的作用。Objective : To study the effect of enhancing curative effect and reducing side-effect on 6-hydroxydopamine induced Parkinson's disease model in rats by Xingnao Chuchan Decoction. Methods : The SD rat brain located in the medial forebrain bundle ( MFB )parts injected 6-hydroxydopamine ( 24 μg ) made for PD model rats. The successful PD rats were randomly divided into 5 groups ( each group of 10 eases, only male and female half ), namely model group, the madopar group ( 75 mg/kg ), levodopa group ( 60 mg/kg ), beta-asarone group ( 15 mg/kg ), Xingnao Chuehan Decoction group ( levodopa 60 mg/kg + beta-asarone 15 mg/kg ). Dosage groups according to the above dose lavage, 2 times a day, for 30 days. End of dosing observed between groups within five minutes of AIM score ( mouth face, axial and forelimbs ), calculating coefficient of liver and kidney and HE staining to observe the striatmn tissue pathological morphology. Resuhs : Compared with control group, model group and the mouth of the face, axial and forelimbs AIM score were significantly increased ( P〈0.01 ), liver and kidney coefficient of no significant difference (P〉0.05), nerve cells in the striatum was less, the residual nucleus pyenosis neurons, part of the cell cytoplasm markedly swollen, suggesting that the PD rat made success and 6-hydroxy dopamine die agent on the model of rat liver and kidney toxicity. Compared with model group, AIM score of every dose group were significantly reduced (P〈0.01), number of striatal neurons after drug treatment was slightly increased, structure the basic normal, no obvious inflammatory infiltration phenomenon of degeneration and necrosis, which the kidney coefficient of levodopa group increased significantly ( P〈0.05 ). Compared with levodopa group, the dobutamine group and the refreshing defibrillation party group of the mouth and face, axial and forelimbs AIM score and the right kidney coefficient were significantly reduced ( P〈0.05 ).
