摘 要:目的:通过建立不同病机引起的两种湿热模型,探索湿热证的肠道菌群组成变化规律。方法:将BALB/c小鼠分成正常组、LPS组、模型A组、模型B组4组;模型A、B组给于高蛋白饲料饲养、20%蔗糖水饮用,置于设定温度为(35±0.15)℃,湿度为80%~90%的人工气候箱中模拟外湿热环境,模型A组给于"花生油脂+白酒"灌胃、模型B组给于"20%番泻叶药液+白酒"灌胃42 d致脾虚;于造模56 d LPS组、模型A、B组给于1mg/kg剂量的脂多糖LPS工作液腹腔注射完成湿热模型造模,于注射LPS工作液后6 h取材进行相关指标检测。结果:模型A、B组均出现了湿热证类似的临床表现、体征和相关炎症因子的改变,干扰素γ(INF-γ)、肿瘤坏死因子α(TNF-α)水平均出现升高;两种湿热模型肠道菌群大肠杆菌属、肠球菌属、梭菌属均出现不同程度的含量增加;双歧杆菌属、乳杆菌属在模型A组中含量增加而在模型B组中含量减少。结论:温病湿热证存在肠道菌群结构失调,湿热证模型出现大肠杆菌属、肠球菌属、梭菌属等条件致病菌过度增长;而双歧杆菌属、乳杆菌属等益生菌的含量受湿热发病机制的影响出现含量表达的差异,脾阳虚为内湿基础的湿热证双歧杆菌属、乳杆菌属含量减少,湿邪困扰为内湿基础的湿热证双歧杆菌属、乳杆菌属含量增多。Objective: Our aim was to explore composition of intestinal flora about febrile disease damp-heat syndrom by establishing two kinds of febrile disease damp-heat syndrome mice models. Methods: Forty mice were divided into four groups: the normal control group,the lipopolysaccharide group,the A model group and the B model group. The A and B model groups wre given high protein food,20% sugar water and were put into the artificial temperature chamber at 35℃,80% ~ 90% air humidity for 12 h every day for 56 days. The A model group was given peanut oil + rice wine by gavage for 42 days and the B model group was given 20% Folium Sennae + rice wine by gavage for 42 days. On the 56 th day,the lipopolysaccharide group,the A and the B model groups were given 1 mg/kg lipopolysaccharide by intraperitoneal injection and then the febrile disease damp-heat syndrome was finished. Results: The two kinds of models showed similar clinical symptoms of febrile disease damp-heat syndrome. The level of interferon( INF-γ) and tumor necrosis factor( TNF-α) increased in two kind of models. Levels of Escherichia,enterococcus and Clostridium increased in two kind of models and levels of Bifidobacterium and Lactobacillus increased in the A model but decreased in the B model. Conclusions: This is to express the imbalance of composition of intestinal flora in febrile disease damp-heat syndrome. Two kinds of models show that levels of pathogenic bacteria was overgrowth,such as Escherichia,enterococcus and Clostridium. Levels of Probiotics expresses different results between the A and the B models. On the basis of spleen Yang deficiency,levels of Bifidobacterium and Lactobacillus are decreased. On the basis of damp stasis,levels of Bifidobacterium and Lactobacillus is increasing.
