摘 要:方法:针对大肠癌围手术期患者的主要中医证候阴虚证,采集符合条件病例的大肠癌组织及癌周正常组织,采用罗氏公司含12×135K个基因的人全基因组芯片检测大肠癌组织差异表达基因,筛选并分析阴虚证癌组织组和癌周正常组织组的差异表达基因,利用Pathway对相关基因进行生物信息学分析。结果:得出大肠癌围手术期患者阴虚证癌组织组和癌周正常组织组表达的差异基因共4869个,包括上调基因1574个,下调基因3295个。经检索数据库Gene Bank,获取基因名称等相关信息。进行Pathway分析,筛选出阴虚证癌组织组和癌周正常组织组之间具有显著差异表达基因主要涉及的信号转导通路。结论:大肠癌围手术期阴虚证癌组织组与癌周正常组织组比较,存在组织差异基因表达特征图谱,主要涉及主要涉及炎症反应,细胞内吞功能,矿物质吸收,酮体合成与分解功能,免疫功能等相关基因。其主要信号转导通路包括MAPK信号通路及p53信号通路等。Objective: A total of 12x135 K human genome microarray was used to detect the expression of differentially expressed genes in colorectal cancer tissues and peritumoral tissues of patients with colorectal cancer. To compare the differentially expressed genes between Yin deficiency syndrome group and peritumoral normal group and analyze the related genes by bioinformatics. Methods: There were 4869 differentially expressed genes in peri-operative patients with Yin deficiency syndrome and normal peritumoral tissues,of which 1574 were up-regulated and 3295 down-regulated. After searching the Internet biology public database Gene Bank,the name of each gene and other basic information were obtained. Pathway analysis was carried out to screen out the signal transduction pathways involved in the differential gene expression between tumor-bearing and normal peritumoral groups. Conclusion: Compared with normal peritumoral masses,there were different gene expression profiles of the genes involved in inflammation,cell endocytosis,mineral absorption,synthesis and decomposition of ketone bodies,immunological function and other related genes in colorectal cancer during perioperative period. The main signal transduction pathways include MAPK signal pathway and p53 signal pathway.
