摘  要:目的:研究唑来膦酸(Zoledronate acid)和异戊烯焦磷酸(IPP)体外扩增前列腺癌患者外周血中γδ T细胞的效率。方法:密度梯度离心法分离外周血中单个核细胞(PBMCs),分别加入5.0μmol/L Zol或10.0μg/ml IPP联合1 000 IU/ml的rh IL-2于含10%自体血浆的AIM-V培养体系培养,观察并收集细胞,采用流式细胞仪检测刺激后CD3^+Vγ^(9+)的γδ T细胞数量和比例,并计算其扩增效率。结果:经14天扩增培养后,两种磷酸抗原均能在体外扩增前列腺癌患者外周血中的γδ T细胞,Zol组的γδ T细胞在淋巴细胞中最高含量达64.7%,IPP组最高达55.9%,且扩增培养至第9天的γδ T细胞对肿瘤细胞有一定的杀伤活性,但Zol组的扩增效率以及细胞的杀伤活性均明显强于IPP组(P<0.05)。结论:Zol具有与IPP体外扩增外周血中γδ T细胞的能力,Zol刺激法可能成为体外扩增前列腺癌患者外周血中γδ T细胞更为经济安全的选择。Objective: To compare the efficiency of Zoledronate acid (Zol) and isopentenyl pyrophosphate (IPP) to stimulate γδ T cell expansion from prostate cancer patient s peripheral blood. Methods : Peripheral blood mononu-clear cells (PBMCs) were isolated by Ficoll - Paque gradient centrifugation, and then cultured in AIM - V medium supplemented with 10% autologous serum,then add 5.0μmol/L Zol or 10.0μg/ml IPP co - cultrue with rhIL -2.The cells were observed and collected and analyzing proportion and total cell amount of CD3 ^+ Yγ9^ + T cells in total PBMCs by using flow cytometry approach. Results : After 14 days culturing, both of the two antigens could amplify yh T cells in peripheral blood of prostate cancer patient in vitro. The group cultured with the medium supplemented with 5. 0 μmol/L Zol plus 1 000 IU/ml IL -2 shown highest content of 64. 7% ,the group cultured with the medium sup-plemented with 10. 0μg/ml IPP plus 1 000 IU/ml IL - 2 shown highest content of 55.9% . γδ T cells cultured in the 9th day showed optimal tumor cells killing activities. But the efficiency of Zol group and cell killing activity were sig-nificantly stronger than IPP group (P 〈0. 05). Conclusion: Both Zol and IPP had the ability to amplify γδ T cells in vitro. But the Zol might be a more economical and safe option for the expansion of γδ T cells in peripheral blood of prostate cancer patient in vitro.
