摘  要:目的观察青蒿琥酯(artesunate,Art)对人Tenon囊成纤维(human Tenon’s capsule fibroblasts,HTFs)细胞增殖与凋亡的影响,探讨青光眼术后滤过泡瘢痕化的治疗对策。方法体外培养HTFs细胞,应用不同浓度(50μg·mL^(-1)、100μg·mL^(-1)、150μg·mL^(-1)、200μg·mL^(-1))的Art处理细胞,采用MTT法检测Art对细胞增殖的影响,流式细胞仪检测Art对细胞凋亡的影响,Western blot检测Art对凋亡相关蛋白Bax、Bcl-2表达的影响。结果 Art作用细胞48 h后,MTT结果显示:与空白对照组比较,不同浓度的Art处理组对HTFs细胞的增殖均有抑制作用,且呈浓度依赖性抑制(均为P<0.05)。流式细胞仪结果显示:与空白对照组比较,50μg·mL^(-1)、100μg·mL^(-1)、150μg·mL^(-1)、200μg·mL^(-1) Art作用后细胞凋亡率逐渐增加(均为P<0.05),分别为8.80%±0.88%、11.60%±0.56%、16.30%±1.03%、23.40%±1.62%。Western blot检测结果显示:与空白对照组比较,不同浓度Art处理组Bax蛋白表达明显增加,Bcl-2蛋白表达明显降低,且均呈浓度依赖性(均为P<0.05)。结论 Art可抑制HTFs细胞增殖并诱导其凋亡,其机制可能是通过调节凋亡相关蛋白Bax、Bcl-2的表达而实现的。Art可能成为一种潜在的抗青光眼术后滤过泡瘢痕化的药物。Objective To investigate the effects of artesunate (Art) on cell pro- lfferation and apoptosis of human Tenon' s capsule fibroblasts ( HTFs), and discuss the countermeasures of bleb sca.rring in glaucoma. Methods In vitro, HTFs were cultivat- ed and applicated by different concentrations (50 μg mL-1, 100 μg mL-1, 150 μg mL-1 ,200 μg mL-1 ) of Art for 48 hours. The effect of Art on cell proliferation was as- sessed by MTT method. The rate of apoptosis induced by Art was determined by flow cytometry. Western Blot was performed to detect the relative expression levels of Bax and Bcl-2 after Art was treated. Results After treated with Art for 48 hours, com- pared with blank control group,Art (50 μg mL-1 , 100 μg mL-1, 150 μg mL-1 ,200μg mL-1) group exhibited notable anti-proliferative effect on HTFs with concentra- tion-dependence ( all P 〈 0.05 ). The results of flow cytometry showed that the apoptosis rates (8.80% ±0.88% ,11.60% ±0.55% ,15.30% ±1.03% ,23.40% ±1.52% ) of HTFs were significantly enhanced with the increase of Art concentration ( all P 〈 0.05 ). The relative expression levels of Bax were obviously high with the increase of Art concentra- tion, while Bcl-2 levels were significantly low with the increase of Art concentration ( all P 〈 0.05). Conelnsion Art can inhibit the proliferation and induce cell apoptosis of HTFs possibly by enhancing the expression of Bax and reducing the expression of Bcl- 2. Art may be a potential drug in preventing fibrous scar formation after glaucoma filtra- tion surgery.
