摘  要:目的构建一种能确切反映脑性瘫痪发生后生理病理改变的动物模型,评价Morris水迷宫行为学试验对检测脑性瘫痪动物模型的适用性。方法随机将30只7日龄SD大鼠分为两组:脑瘫模型组20只,对照组10只。两组实验动物均给予行为学观察(不自主运动、悬吊、斜坡、倾斜板等试验与肌张力测定)及Morris水迷宫实验。结果不自主运动测试脑瘫模型组为(9.50±0.89)分,对照组为(10.00±0.00)分;悬吊测试中,脑瘫模型组与对照组的时间分别为(2.03±0.79)分和(4.15±0.67)分;斜坡测试中,脑瘫模型组与对照组的时间分别为(11.25±1.21)秒和(4.35±0.93)秒;肌张力测试脑瘫模型组为(7.80±0.62)分,对照组为(10.00±0.00)分,以上各试验结果两组比较差异均有统计学意义(P<0.05)。在倾斜板试验中,脑瘫模型组均出现阳性表现,对照组无阳性反应。Morris水迷宫实验中,脑瘫模型组的逃避潜伏期在各个时间点的差异均有统计学意义(P<0.05)。结论 7日龄新生大鼠单侧颈总动脉结扎可成功构建痉挛型脑瘫动物模型,脑损伤的部位主要在白质,脑瘫模型体征典型。与传统评价方法比较,Morris水迷宫行为学试验对鉴定脑瘫模型同样适用可靠。Objective To estimate an animal model of spastic cerebral palsy (CP) exactly reflected the physiological and pathological changes after the occurrence of cerebral palsy and evaluate the applicability of behavioral tests in the detection of spastic cerebral palsy model. Methods 30 7-day-old rats were randomly divided Spastic cerebral palsy model group(20 rats) and control group (10 rats). The behavioral changes of rats were detected by suspension test, slope test, muscle tension test, inclined plate test, non self movement observation, and Morris water maze test. Results The score of suspension test of experimental group and control group were 2.03 ± 0.79 and 4.15 ± 0. 67 (P〈0. 05). The time of slope test of experimental group and control group were 11.25 ± 1.21 seconds and 4.35 ± 0.93 seconds (P〈0.05). The score of muscle tension test of experimental group and control group were 7.80 + 0.62 and 10. 00 + 0. 00 (P〈 0.05). The score of non self movement of experimental group and control group 9.50 + 0.89 and 10. 00 + 0. 00 (P〈 0.05). The inclined plate test of experimental group was positive. The inclined plate test of control group was negative. There were statistically significant differences in the escape latency of experimental group and control group at different time points in Morris water maze test. Conclusion Model of spastic cerebral palsy in rats maintains typical signs and symptoms for long time. The operation is simple and standardized. Behavioral experiments are reliable applications on the identification of spastic cerebral palsy animal model.
