摘　　要：目的研究PHLPP2在人正常脑组织以及脑胶质瘤组织中的表达及其临床意义。方法选取青岛大学附属医院与安丘人民医院神经外科自2014年12月至2016年10月间手术切除并经病理证实的人脑胶质瘤标本56例,其中I级7例,Ⅱ级19例,Ⅲ级21例,胶质母细胞瘤9例。另取20例因脑创伤行内减压术患者的正常脑组织标本作为对照。应用RT-PCR、Western-blotting分别检测各标本中PHLPP2m RNA和PHLPP2蛋白的表达。结果 RT-PCR和Western-blotting检测结果显示正常脑组织中PHLPP2 mRNA和PHLPP2蛋白表达最高,不同级别胶质瘤组织PHLPP2 mRNA和PHLPP2蛋白表达差异有统计学意义(P<0.05),且随着肿瘤病理级别的增高,胶质瘤组织中PHLPP2 mRNA和PHLPP2蛋白表达值降低,差异有统计学意义(P<0.05)。结论 PHLPP2在正常脑组织和不同级别的脑胶质瘤组织中都有表达,其表达值与肿瘤的病理分级呈负相关,PHLPP2可能作为一个肿瘤抑制基因在胶质瘤的发生、发展中起到关键性的作用。Objective To investigate the expression of PHLPP2 in the tumor tissues of patients with gliomas and evaluate its clinical significance. Methods Fifty-six patients with brain gliomas（7 with grade Ⅰ,19 grade Ⅱ,21 grade Ⅲ and 9 with glioblastoma）performed surgical excision in Our hospitals from December 2014 to October 2016 were chosen and 20 case of normal brain tissues from patients performed decompression operation resulting from cerebral hernia were selected as the controls in our experiment.RT-PCR and Western-blotting were used to detect the mRNA and protein expressions of PHLPP2. Results The results investigated by RT-PCR and Westem-blotting were consistent, the highest expression of PHLPP2 was in the normal brain tissues. The expression value between different grades of tumors was obviously significant and decreased in a grade-dependent manner（P〈0.05）. Conclusion The mRNA and protein expressions of PHLPP2 are noted with positive correlation to the pathological grades of the gliomas, indicating that PHLPP2 may play an important role in the generation and development of gliomas.
