摘 要:目的观察PI3K-Akt信号通路在尼古丁抑制小胶质细胞增殖中的作用。方法培养的小胶质细胞系(BV-2)中分别加入不同浓度的尼古丁(Ni)、谷氨酸(GLU),分为谷氨酸组、尼古丁组以及谷氨酸和尼古丁共同添加组,培养24h,采用MTT方法检测小胶质细胞存活率,Western Blot方法检测PI3K相关蛋白(P85)的表达情况。结果通过MTT检测发现:(1)500μmol/L浓度的谷氨酸促进小胶质细胞OD值增高(2)尼古丁能够使小胶质细胞OD值降低(3)不同浓度的尼古丁能够抑制500μmol/L谷氨酸诱导的小胶质细胞OD值的升高,10μmol/L效果最显著。通过Western Blot实验观察到:(1)谷氨酸(浓度500μmol/L)使小胶质细胞PI3K-Akt信号通路的P85蛋白表达增多(2)尼古丁(浓度10μmol/L)抑制小胶质细胞PI3KAkt信号通路的P85蛋白表达(3)在尼古丁存在的条件下,谷氨酸未能使小胶质细胞PI3K-Akt信号通路的P85蛋白表达增多。结论 (1)尼古丁可以抑制谷氨酸诱导的小胶质细胞的增殖(2)PI3K-AKT信号通路参与尼古丁抑制小胶质的细胞增殖。Objective To investigate the role of PI3K - Akt signaling pathway in the inhibition of microglia proliferation by nicotine. Methods Different concentrations of nicotine (Ni) and glutamate (GLU) were added into cultured microglia cell line ( BV - 2 ), culture 24 hours, cell viability was detected by MTT assay, observe the effect of the number of microglia, the expression of PI3 K/Akt related protein ( P85 ) was detected by Western Blot. Results MTr results show: ( 1 ) 500μmol/L concentration of glutamate promoted the increase of OD (2) The concentration of nicotine could decrease the OD of microglia (3) Different concentration of nicotine can inhibit the increased OD of microglial which induced by glutamate. Western Blot results show: ( 1 ) The concentration of 500μmol/L glutamate increased the protein expression of P85 in the PI3 K -Akt signaling pathway of microglia (2) The concentration of 10μmol/L nicotine decreased the protein expression of P85 in the PI3K - Akt signaling pathway of microglia (3) In the presence of nicotine, glutamate failed to increase the expression of P85 protein in the PI3K - Akt signaling pathway of microglia. Conclusion ( 1 ) Nicotine can inhibit the proliferation of microglia induced by glutamate (2) PI3K- AKT signaling pathway is involved in the inhibition of microglia proliferation by nicotine.
