摘 要:目的通过下调脑挫伤细胞中laminin,观察其对新生大鼠皮质神经干细胞迁移能力的影响。方法设计合成下调大鼠laminin的siRNA并转染至大鼠脑挫伤细胞,realtime PCR检测脑挫伤细胞中laminin基因水平siRNA干扰效率;westernblot检测脑挫伤细胞中laminin蛋白水平siRNA干扰效率;迁移实验检测神经干细胞迁移率。结果大鼠脑挫伤细胞转染下调laminin的siRNA后,laminin基因表达水平下降(71.35±4.26)%,laminin蛋白水平较对照组下降(50.1±3.45)%;迁移实验显示siRNA组神经干细胞迁移能力下降,P<0.05。结论下调脑挫伤细胞中laminin表达,可以抑制神经干细胞迁移能力,因此尽量保存脑外伤中挫伤组织有利于损伤区域神经再生。Objective To study the effect of laminin on the migration of cortical neural stem cells in neonatal rats by downregulating laminin in cells of TBI. Methods Design and syn- thesis the siRNAs for laminin ,and transfected into cells of TBI. The efficiency of siRNA interfer- ence were detected by realtime PCR and westernbiot . Neural stem cell migration assay were ap- plied to observe NSC migration rate in each group. Results The laminin gene expression level of cells of TBI decreased (71.35±4.26)% ,compared with negetive control group;The laminin pro- tein expression level of cells of TBI decreased(50.1±3.45)%, compared with negetive control group;and migration assay showed that the neural stem cell migration ability decreased in siRNA group ,compared with control group,P〈0.05. Conclusion Downregulation of laminin expression in cells of TBI can inhibit the migration of neural stem cells,so it is beneficial to preserve the in- jured tissue of brain injury patients for the neuroregeneration of injured area.
