摘 要:研究高F值玉米寡肽-锌螯合物的分离纯化及结构.利用膜分离系统、葡聚糖凝胶G-15和C-18反相高效液相对高F值玉米寡肽-锌螯合物进行分离纯化,运用紫外吸收光谱、电喷雾电离质谱争红外吸收光谱研究螯合物的结构.紫外全波长扫描结果显示,硫酸锌的最大吸收波长在192nm,高F值玉米寡肤的最大吸收波长在220nm,高F值玉米寡肽-锌螯合物的最大吸收波长在209nm;经过分离纯化高F值玉米寡肽-锌螯合物,得到组分P-Zn3a和P-Zn3b;采用MALDI-TOE/TOF MS/MS对组分进行测序,氨基酸序列为甘氨酸-缬氨酸-亮氨酸(异亮氨酸)(Gly-Val-Leu(Ⅱe))和甲硫氨酸-亮氨酸(异亮氨酸)(MetLeu(Ⅱe));红外光谱结构表征显示玉米寡肽与锌离子螯合机制为:与Zn^+发生螯合的位置可能是-NH-C=O上的-NH,-COOH上的-OH和末端-NH2.由此可初步推断,高F值玉米寡肽与锌发生了螯合并产生了一种新型的螯合物.Objective:To study the purification and structure of zinc chelate with high F value of corn oli- gopeptide. Methods: The zinc chelate with high F value of corn oligopeptide was isolated and purified by Mem- brane Separation System, Sephadex G - 15 and C - 18 reversed - phase high performance liquid chromatogra- phy. The structure of the chelate was studied by UV absorption spectroscopy, infrared absorption spectroscopy and electrospray ionization mass spectrometry. Results:Uv wavelength scanning all the results show that the maximum absorption wavelength of zinc sulfate in 192 nm, the maximum absorption wavelength of corn oli- gopeptides in 220 nm, the maximum absorption wavelength of peptide zinc chelate at 209 nm. The components P -Zn3a and P -Zn3b are acquired. The component are sequenced with MALDI -TOF/TOF MS/MS, and the amino acid sequence are Gly -Val -Leu(Ile) and Met -Leu(Ile). The infrared spectrum of the structure indicated that the chelating mechanism of corn oligopeptide and zinc ion was as follows: the position of chela- tion with Zn2 + may be - NH on - NH - C = O of oligopeptide, while - OH on - COOH and terminal - NH2 of oligopeptide. Conclusion : It can be inferred that high F value of tom oligopeptides chela merger with zinc produced a new type of chelate.
