摘  要:埃博霉素是纤维堆囊菌产生的一种和紫杉醇具有相同抗癌作用的大环内酯类次级代谢产物。以So ce2161为出发菌株,通过一系列单因素实验确定了对产量影响最大的四个因素,即温度、初始pH、转速、接种量。对四个因素设计正交实验进行发酵条件优化,最终得到最佳实验条件为使用250 m L挡板三角瓶,在温度32℃、初始pH=7.2、转速180 r/min、接种量6%、装液量60 m L/250 m L、发酵时间6 d、种龄60 h条件下,埃博霉素B的产量为46.83 mg/L。The Epothliones,compounds with anticancer mechanisms similar to that of paclitaxel ,are the secondary metabolites which are produced by the Soranguim cellulosum.Four factors such as temperature,initial pH,moving speed and inoculum concentration were determined by a series of single-factor experiment.In this study,the fermentation conditions will be optimized by the single-factor experiment and orthogonal experiment,in order to get the most suitable experimental conditions.The following optimal experimental conditions are used:Sorangium cells were grown in the suitable medium in a 250 mL Erlenmeyer flask with the So ce2161 volume of 6%.Cells were grown at 32 ℃.Liquid cultures were shaken at 180 rpm.The medium adjusted to pH=7.2.The strain age of the So ce2161 is 60 h.The production of Epothlione B is 46.83 mg/L after 6 days.
