摘 要:为研究不同鸡种骨骼肌卫星细胞生肌决定因子5(Myf5)基因表达差异,试验通过提取大围山微型鸡和艾维茵肉鸡骨骼肌卫星细胞并进行两个鸡种骨骼肌卫星细胞增殖分化差异及Myf5基因表达差异研究。结果显示:两个鸡种骨骼肌卫星细胞生长曲线呈"S"形,大围山微型鸡骨骼肌卫星细胞生长速度较低;在分化过程中,艾维茵肉鸡骨骼肌卫星细胞分化能力高于大围山微型鸡,且艾维茵肉鸡骨骼肌卫星细胞的Myf5 mRNA表达量显著高于大围山微型鸡(P<0.05)。本试验表明:Myf5基因是影响家禽肌肉生长发育的重要候选基因,且细胞体外培养对家禽分子育种研究具有一定科学指导意义。In order to study Mff5 gene expression in skeletal muscle satellite cells of different chicken breeds, Daweishan mini chickens and Avian broilers were selected to study the differentiation and MyfS gene expression of skeletal muscle satellite cells. The results showed that the growth curves of skeletal muscle satellite cells from the two chicken breeds presented "S", and growth rates of skeletal muscle satellite cells from Daweishan mini chickens were lower than that in Avian broilers. In the process of differentiation,Vvian broilers showed higher skeletal muscle satellite cell differentiation ability than Daweishan mini chickens, as well as Myf5 gene mRNA expression in skeletal muscle satellite cells of avian broilers was significantly higher than that in Daweishan mini chickens (P〈0.05). The results suggested that Myf5 gene was an important candidate gene for the development of muscle in poultry, and cell culture in vitro showed scientific guiding significance on study of poultry molecular breeding.
