摘 要:目的:探讨口腔鳞癌中的核基质蛋白特异AT序列结合蛋白2(SATB2)表达水平的临床意义和对口腔鳞癌生物学行为的影响。方法:收集58例口腔鳞癌标本,检测SATB2的表达情况,分析SATB2表达水平与病理参数之间的关系及与患者总体生存率的关系;用载有SATB2的慢病毒转染HN6细胞,Western blot及实时定量RT-PCR检测转染效果,并通过划痕愈合、Transwell、Invasion实验检测SATB2对HN6细胞生物学行为的影响。结果:统计学结果表明,高表达的SATB2与患者年龄、性别、病理分级无关,但与TNM分期、颈淋巴结转移及临床分期密切相关,与患者的不良预后密切相关;SATB2成功转染HN6细胞系并得到稳定过表达SATB2的Lv-SATB2-HN6细胞;过表达SATB2的HN6细胞迁移、侵袭能力明显高于HN6细胞。结论:SATB2在口腔鳞癌组织及细胞系中表达增高,过表达SATB2能促进肿瘤细胞迁移、侵袭能力。Objective : To investigate the expression of nuclear matrix protein- special AT-rich sequence binding protein 2 (SATB2) in oral squamous cell carcinoma (OSCC) and its effect on the biological behavior of oral squamous cell carcinoma. Methods : 58 cases of oral squamous cell carcinoma were collected and the expression of SATB2 was detected by immunohistochemical staining. The rela-tionship between SATB2 and the pathological parameters and the overall sur^^ival rate was analyzed. Then the HN6 cell lines were trans-fected with lentivirus containing SATB2 and tested through western blot and RT-PCR as well. Wound healing test,migration tests and invasion tests were preformed for further study of biological roles of SATB2 in HN6 cell lines. Results : The high expression of SATB2 was not associated with age, sex and pathological grade, but was closely related to TNM stage, cervical lymph node metastasis and clin-ical stage, which was closely related to the poor prognosis. The results of Western blot and PCR showed that SATB2 was successfully transfected into HN6 cell line and Lv-SATB2-HN6 cells stably overexpressing SATB2 were obtained. The HN6 cells overexpressing SATB2 migrated and the invasive ability was significantly higher than that of HN6 cells. Conclusions : The expression of SATB2 in oral squamous cell carcinoma and cell line increased, and overexpression of SATB2 could promote the migration and invasion of tumor cells.
