转录共刺激因子对结直肠癌基质金属蛋白酶及相关蛋白的影响

摘　　要：目的:探讨转录共刺激因子(TAZ)对结直肠癌基质金属蛋白酶及相关增殖蛋白的影响,分析其影响结直肠癌肿瘤生物学行为的可能作用机制。方法:采用合成TAZ小干扰RNA(TAZ-siRNA)转染结直肠癌细胞株HCTT116抑制TAZ表达,检测细胞活性、侵袭迁移能力、细胞凋亡,采用实量定量反转录聚合酶链反应(RT-qPCR)检测TAZ与基质金属蛋白酶mRNA表达,采用Western blot法相关蛋白表达。结果:转染组TAZ mRNA与蛋白表达均明显低于对照组与空白组(t=27.116、17.600、24.051、16.168,P<0.01);HCT116细胞活性、侵袭迁移能力均明显低于对照组与空白组,细胞凋亡率均明显高于对照组与空白组(t=12.944、17.613、20.489、12.639、18.324、20.051,P<0.01);基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)等mRNA表达均明显低于对照组与空白组,金属蛋白酶抑制因子-1(TIMP-1)mRNA表达均明显高于对照组与空白组(t=13.588、10.041、6.678、13.164、10.479、6.705,P<0.05或<0.01);B细胞淋巴瘤/白血病-2(bcl-2)蛋白表达均明显低于对照组与空白组,bcl-2相关X蛋白(bax)、细胞色素C(Cyt C)表达均明显高于对照组与空白组(t=9.358、7.670、15.205、19.163、5.937、17.063,P<0.05或<0.01)。结论:抑制转录共刺激因子(TAZ)能够抑制结直肠癌肿瘤细胞增殖与转移,可能与调节基质金属蛋白酶、相关蛋白表达水平有关。Objective： To study the effect of matrix metalloprotein and proliferin of TAZ for coloreetal cancer, and analyzed possible mechanisms of colorectal cancer biological behavior.Method： Using TAZ-siRNA transfection colorectal cancer cell line HCTr116 inhibits TAZ expression, and cell activity, attack mobility, apoptosis, TAZ and matrix metal protease mRNA expression, related protein expression were tested.Result： Transfection groups TAZ mRNA and protein expression were significantly lower than those of the control group and the blank group （t=27.116, 17.600, 24.051, 16.168; P〈0.01 ） .HCT116 cell activity, attack mobility ability were significantly lower than those of the control group and the blank group, apoptosis rate was significantly higher than those of the control group and the blank group （t=12.944, 17.613, 20.489, 12.639, 18.324, 20.051; P〈0.01 ） .MMP-2, MMP-9 mRNA expression were significantly lower than those of the control group and the blank group, TIMP-1 mRNA expression was significandy higher than those of the control group and the blank group （ t=13.588, 10.041, 6.678, 13.164, 10.479, 6.705; P〈0.05 or〈O.01 ） .Bcl-2 protein expression was significantly lower than those of the control group and the blank group, bax, Cyt C expression were significantly higher than those of the control group and the blank group （t=9.358, 7.670, 15.205, 19.163, 5.937, 17.063; P〈0.05 or 〈0.01 ） .Conclusion： TAZ can inhibit tumor cell proliferation and metastasis of colorectal cancer, it may be related to regulating the substrate metal protease and related protein expression levels.
