摘  要:目的研究不同浓度绿原酸对肝癌细胞的抑制作用,以及对肝癌细胞MAPK/ERK信号通路的影响。方法分别应用50,125,250,500,1 000μmol/L绿原酸与肝癌细胞株HepG_2细胞共孵育24 h,Trypan blue染色并进行HepG_2细胞计数,Western blotting测定HepG_2细胞MAPK/ERK信号通路的ERK1/2磷酸化,CM-H_2DCFDA荧光探针测定HepG_2细胞内ROS浓度,观察绿原酸对HepG_2细胞增殖的抑制作用。结果与0μmol/L组比较,500,1 000μmol/L组HepG_2细胞的增殖分别降低了45.5和81.7个百分点;绿原酸抑制HepG_2细胞MAPK/ERK信号通路的ERK1/2磷酸化(P<0.01);绿原酸促进HepG_2细胞内ROS浓度升高。结论绿原酸通过抑制MAPK/ERK信号通路的活化遏制肝癌细胞增殖。Objective To investigate the effect of chlorogenic acid (CGA) on hepatocarcinoma cells and MAPK/ERK pathway. Methods HepG2 cells were co-incubated with 50,125,250,500,1 000 μmol/L CGA for 24 h respectively. HepG2 cells were counted after Trypan blue staining. ERK1/2 phosphorylation of MAPK/ERK pathway was detected by Western blotting. Intracellular concentra- tion of reactive oxygen species(ROS) was detected using fluorescent probes after treatment with CGA. Results Compared with blank group,the proliferation of HepG2 ceils increased by 45. 5 and 81.7 percentages in 500, 1000 μmol/L CGA group. The 500, 1 000 μmol/L CGA inhibited ERK1/2 phosphorylation( P 〈 0. 01 ). Fluorescent probes results showed that CGA promoted ROS regeneration in HepG2 cells. Conclusion CGA may inhibit the proliferation of hepatoearcinoma cells through preventing the activation of MAPK/ ERK pathway.
