摘  要:目的探讨乳腺癌组织芯片HER2 FISH检测结果的可靠性。方法收集192例乳腺癌病例,制成直径0.9 mm的组织芯片,进行HER2 FISH检测,对照常规石蜡切片的检测结果,进行统计学分析。结果组织芯片中HER2 FISH的阳性率为25.0%(48/192),常规石蜡切片的阳性率为26.0%(50/192),配对χ~2检验显示两组之间差异无统计学意义(P>0.05)。有4例乳腺癌病例存在检测结果的不一致,其中常规石蜡切片检测为点状扩增病例中有3例组织芯片检测为阴性,常规石蜡切片检测为无扩增病例中有1例组织芯片检测为点状扩增。结论组织芯片技术在乳腺癌HER2 FISH检测中的可靠性强,但对于临床中乳腺癌病例建议使用常规石蜡切片进行HER2 FISH检测。Objective To explore the reliability of tissue chip in the research of human breast cancer HER2 FISH detection. Methods Tissue microarray(TMA) with 0. 9 mm in diameter tissue core was constructed from 192 cases of human breast cancer. HER2 was de- tected by FISH technique. At the same time the results of the tissue microarrays were compared with those of the routine paraffin section. Results The positive rates in HER2 FISH detection of breast cancer were 25.0% and 26.0% by TMA and the routine paraffin sec- tion,respectively, and there was no statistical difference (P 〉 0. 05). But the results of 4 cases were different between the two methods. Three cases of dot expansion using the routine paraffin section were negative by TMA, and 1 case of dot expansion using TMA showed no expansion by the routine paraffin section. Conclusion Tissue microarrays could be a reliable tool in the application of human breast cancer HER2 FISH detection,but it is better to use the routine paraffin section in the clinical.
