摘  要:目的:探讨肢体缺血后处理在脑缺血再灌注中激活自噬最佳时间窗。方法:用线栓法制备大鼠局灶性脑缺血再灌注模型,于大鼠脑缺血2 h再灌注24 h后进行神经功能缺损评分、脑梗死体积测定及HE染色、免疫组化检测自噬相关蛋白LC3-Ⅱ。结果:与假手术组(Sham)比较,缺血再灌注组(I/R)出现明显神经功能缺损症状及脑梗死改变,LC3-Ⅱ表达不明显,与I/R组比较,肢体缺血后处理组(Lpost C)神经功能缺损症状有所改善,脑梗死体积显著缩小,LC3-Ⅱ表达明显增加,且在Lpost C0min亚组增加最显著(P<0.05)。结论:肢体缺血后处理对脑缺血再灌注损伤有保护作用,并且再灌注即刻干预激活自噬最显著。Objetive: To investigate the optimal time window to activate autophagy after limb ischemia postconditioning in cerebral ischemia reperfusion. Methods: The rat model of focal cerebral ischemia and reperfusion was established by thread embolization. Then,the neurological deficit score,the volume of cerebral infarction,the HE staining were performed and immunohistochemistry were used to detect the autophagy-related protein LC3-Ⅱ 2 h after cerebral ischemia then 24 h after reperfusion. Results:Compared with the sham operated group( Sham),the neurological deficits and cerebral infarction were obvious occured in the ischemia-reperfusion group( I/R),and the expression of LC3-Ⅱ was not obvious.Compared with I/R group,the neurological deficits symptoms in limb ischemic postconditioning group(LpostC) was improved,cerebral infarction volume was significantly reduced,and the expression of LC3-Ⅱ was significantly increased in LpostC 0min subgroup( P〈0.05). Conclusion: Limb ischemic treatment has protective effect on cerebral ischemia reperfusion injury,and immediate reperfusion intervention significantly activates autophagy.
